Nebo se jen hon\u00ed za snem o state\u010dn\u00e9m nov\u00e9m sv\u011bt\u011b, kde se vytv\u00e1\u0159ej\u00ed geneticky modifikovan\u00e9 organismy a implantuj\u00ed do um\u011bl\u00fdch d\u011bloh? Bez ve\u0159ejn\u00e9ho financov\u00e1n\u00ed by se \u017e\u00e1dn\u00fd z t\u011bchto v\u00fdzkum\u016f neuskute\u010dnil, a p\u0159esto se schov\u00e1vaj\u00ed za pr\u00e1zdn\u00e9 fr\u00e1ze o pomoci ne\u0161\u0165astn\u00fdm \u017een\u00e1m, kter\u00e9 nemohou m\u00edt d\u011bti.<\/span><\/em><span dir=\"auto\">\u00a0\u2043 Patrick Wood, editor.<\/span><\/p>\n<p><span dir=\"auto\">V\u011bdci vytvo\u0159ili lidsk\u00e1 embrya odebr\u00e1n\u00edm jader z b\u011b\u017en\u00fdch ko\u017en\u00edch bun\u011bk, jejich vlo\u017een\u00edm do darovan\u00fdch vaj\u00ed\u010dek a jejich oplodn\u011bn\u00edm spermiemi. Tato pr\u00e1ce je laboratorn\u00edm experimentem, kter\u00fd ukazuje, co by nakonec mohlo b\u00fdt mo\u017en\u00e9 u lid\u00ed, kte\u0159\u00ed nejsou schopni produkovat \u017eivotaschopn\u00e1 vaj\u00ed\u010dka, a\u010dkoli je st\u00e1le t\u0159eba p\u0159ekonat zna\u010dn\u00e9 v\u011bdeck\u00e9 p\u0159ek\u00e1\u017eky.<\/span><\/p>\n<p><span dir=\"auto\">T\u00fdm z Oregonsk\u00e9 univerzity zdrav\u00ed a v\u011bdy pou\u017eil techniku \u200b\u200bzvanou p\u0159enos j\u00e1dra somatick\u00fdch bun\u011bk, p\u0159i kter\u00e9 je j\u00e1dro ko\u017en\u00ed bu\u0148ky vlo\u017eeno do darovan\u00e9ho vaj\u00ed\u010dka, ze kter\u00e9ho byl odstran\u011bn vlastn\u00ed genetick\u00fd materi\u00e1l. Po oplodn\u011bn\u00ed spermi\u00ed tyto rekonstruovan\u00e9 bu\u0148ky produkovaly embrya, kter\u00e1 se vyv\u00edjela n\u011bkolik dn\u00ed. A\u010dkoli se jedn\u00e1 o \u010dist\u011b experiment\u00e1ln\u00ed postup, jeho\u017e klinick\u00e9 vyu\u017eit\u00ed je vzd\u00e1leno roky, v\u00fdzkum publikovan\u00fd v \u010dasopise\u00a0<\/span><em><span dir=\"auto\">Nature Communications<\/span><\/em><span dir=\"auto\">\u00a0ukazuje prvn\u00ed d\u016fkazy o\u00a0<\/span><em><span dir=\"auto\">mitomy\u00e1ze<\/span><\/em><span dir=\"auto\">\u00a0, co\u017e je term\u00edn, kter\u00fd t\u00fdm pou\u017e\u00edv\u00e1 pro indukci d\u011blen\u00ed podobn\u00e9ho mei\u00f3ze v somatick\u00e9m genomu, co\u017e by jednoho dne mohlo pomoci \u017een\u00e1m bez funk\u010dn\u00edch vaj\u00ed\u010dek m\u00edt geneticky p\u0159\u00edbuzn\u00e9 d\u011bti.<\/span><\/p>\n<p><span dir=\"auto\">\u201eNeplodnost postihuje miliony lid\u00ed na cel\u00e9m sv\u011bt\u011b a je \u010dasto zp\u016fsobena nedostatkem \u017eivotaschopn\u00fdch gamet,\u201c napsali v\u011bdci. U \u017een se po polovin\u011b t\u0159ic\u00e1t\u00fdch let st\u00e1v\u00e1 hlavn\u00edm faktorem pokles kvality vaj\u00ed\u010dek souvisej\u00edc\u00ed s v\u011bkem a sou\u010dasn\u00e1 l\u00e9\u010dba neplodnosti nem\u016f\u017ee t\u011bm, kter\u00e9 \u017eivotaschopn\u00e1 vaj\u00ed\u010dka zcela chyb\u00ed, pomoci.<\/span><\/p>\n<h3 class=\"wp-block-heading has-medium-font-size\"><strong><span dir=\"auto\">Jak se ko\u017en\u00ed bu\u0148ky staly vej\u010dit\u00fdmi \u00fatvary<\/span><\/strong><\/h3>\n<p><span dir=\"auto\">P\u0159i tomto postupu se j\u00e1dro ko\u017en\u00ed bu\u0148ky implantuje do darovan\u00e9 vaj\u00ed\u010dkov\u00e9 bu\u0148ky, jej\u00ed\u017e vlastn\u00ed genetick\u00fd materi\u00e1l byl odstran\u011bn. Tato technika se naz\u00fdv\u00e1 transfer jader somatick\u00fdch bun\u011bk neboli SCNT. V\u011bdci tuto metodu pou\u017e\u00edvaj\u00ed ke klonov\u00e1n\u00ed zv\u00ed\u0159at po cel\u00e1 desetilet\u00ed, ale t\u00fdm z Oregonu ji upravil pro jin\u00fd \u00fa\u010del: vytvo\u0159en\u00ed rekonstruovan\u00fdch bun\u011bk, kter\u00e9 lze potenci\u00e1ln\u011b oplodnit jako p\u0159irozen\u00e1 vaj\u00ed\u010dka.<\/span><\/p>\n<p><span dir=\"auto\">Kdy\u017e se j\u00e1dro ko\u017en\u00ed bu\u0148ky dostane do prost\u0159ed\u00ed darovan\u00e9ho vaj\u00ed\u010dka, stane se n\u011bco neo\u010dek\u00e1van\u00e9ho. Cytoplazma vaj\u00ed\u010dka, kter\u00e1 zaji\u0161\u0165uje bun\u011b\u010dn\u00e9 mechanismy nezbytn\u00e9 pro d\u011blen\u00ed, nut\u00ed chromozomy ko\u017en\u00ed bu\u0148ky uspo\u0159\u00e1dat se do v\u0159et\u00e9nkov\u00e9 struktury, podobn\u011b jako se to d\u011bje v p\u0159irozen\u00fdch vaj\u00ed\u010dk\u00e1ch. B\u011bhem asi dvou hodin po transferu se viditeln\u00e1 v\u0159et\u00e9nka vytvo\u0159ila u p\u0159ibli\u017en\u011b 77 % rekonstruovan\u00fdch bun\u011bk.<\/span><\/p>\n<p><span dir=\"auto\">K tomu doch\u00e1z\u00ed, i kdy\u017e ko\u017en\u00ed bu\u0148ky nepro\u0161ly replikac\u00ed DNA, kter\u00e1 norm\u00e1ln\u011b p\u0159edch\u00e1z\u00ed bun\u011b\u010dn\u00e9mu d\u011blen\u00ed. P\u0159irozen\u00e9 vaj\u00ed\u010dka obsahuj\u00ed duplikovan\u00e9 chromozomy, z nich\u017e ka\u017ed\u00fd se skl\u00e1d\u00e1 ze dvou spojen\u00fdch kopi\u00ed, naz\u00fdvan\u00fdch sestersk\u00e9 chromatidy. Chromozomy ko\u017en\u00edch bun\u011bk jsou naproti tomu jedin\u00e9, neduplikovan\u00e9 kopie. Cytoplazma d\u00e1rcovsk\u00e9ho vaj\u00ed\u010dka je nicm\u00e9n\u011b indukuje k vytvo\u0159en\u00ed struktury schopn\u00e9 d\u011blen\u00ed.<\/span><\/p>\n<p><span dir=\"auto\">V\u00fdzkumn\u00edci z OHSU p\u0159edstavili novou techniku \u200b\u200bl\u00e9\u010dby neplodnosti, kter\u00e1 p\u0159em\u011b\u0148uje ko\u017en\u00ed bu\u0148ky na vaj\u00ed\u010dka. Na obr\u00e1zku je sn\u00edmek vaj\u00ed\u010dka s jasn\u00fdm obrazem j\u00e1dra ko\u017en\u00ed bu\u0148ky p\u0159ed oplodn\u011bn\u00edm. (Zdroj: Oregon Health &amp; Science University)<\/span><\/p>\n<h3 class=\"wp-block-heading has-medium-font-size\"><strong><span dir=\"auto\">P\u0159ekon\u00e1n\u00ed hlavn\u00ed p\u0159ek\u00e1\u017eky plodnosti<\/span><\/strong><\/h3>\n<p><span dir=\"auto\">Kdy\u017e v\u011bdci k t\u011bmto rekonstruovan\u00fdm bu\u0148k\u00e1m p\u0159idali spermie, v\u011bt\u0161ina z nich z\u016fstala zaseknut\u00e1. P\u0159irozen\u00e9 vaj\u00ed\u010dka v\u011bd\u00ed, jak reagovat na pr\u016fnik spermi\u00ed: Dokon\u010d\u00ed sv\u00e9 d\u011blen\u00ed a p\u0159iprav\u00ed genetick\u00fd materi\u00e1l obou rodi\u010d\u016f ke spojen\u00ed. Ale asi t\u0159i \u010dtvrtiny rekonstruovan\u00fdch bun\u011bk jednodu\u0161e z\u016fstaly na sv\u00e9m m\u00edst\u011b.<\/span><\/p>\n<p><span dir=\"auto\">Um\u011bl\u00e1 aktivace elektrick\u00fdmi impulsy a chemickou l\u00e1tkou zvanou roskovitin \u00fasp\u011b\u0161n\u011b spustila proces d\u011blen\u00ed, co\u017e zp\u016fsobilo, \u017ee bu\u0148ky rozd\u011blily sv\u00e9 chromozomy mezi pronukleus a odlou\u010den\u00e9 pol\u00e1rn\u00ed t\u011bl\u00edsko, podobn\u011b jako se to d\u011bje v p\u0159irozen\u00fdch vaj\u00ed\u010dk\u00e1ch po oplodn\u011bn\u00ed. T\u00edmto p\u0159\u00edstupem p\u0159ibli\u017en\u011b 78 % bun\u011bk odlou\u010dilo pol\u00e1rn\u00ed t\u011bl\u00edsko a p\u0159ibli\u017en\u011b 76 % vytvo\u0159ilo dva pronukley: jeden z chromozom\u016f ko\u017en\u00ed bu\u0148ky a jeden ze spermie.<\/span><\/p>\n<h3 class=\"wp-block-heading has-medium-font-size\"><strong><span dir=\"auto\">Chromozomy n\u00e1hodn\u011b se\u0159azen\u00e9, ne v organizovan\u00fdch p\u00e1rech<\/span><\/strong><\/h3>\n<p><span dir=\"auto\">Aby v\u011bdci sledovali, co se d\u011bje s jednotliv\u00fdmi chromozomy b\u011bhem tohoto neobvykl\u00e9ho procesu, oslovili rodinu s rodi\u010di r\u016fzn\u00e9ho etnick\u00e9ho p\u016fvodu. Pou\u017eili ko\u017en\u00ed bu\u0148ky od dcery a spermie od nep\u0159\u00edbuzn\u00e9ho d\u00e1rce. \u010cten\u00edm sekvenc\u00ed DNA byli schopni ur\u010dit, kter\u00e9 chromozomy poch\u00e1zej\u00ed od matky dcery, kter\u00e9 od jej\u00edho otce a kter\u00e9 od d\u00e1rce spermatu.<\/span><\/p>\n<p><span dir=\"auto\">T\u00fdm vy\u0161et\u0159il 90 embry\u00ed. V p\u0159ibli\u017en\u011b 54 % p\u0159\u00edpad\u016f se chromozomy rozd\u011blily do dvou kompartment\u016f. Zp\u016fsob, jak\u00fdm se d\u011blily, se v\u0161ak v \u017e\u00e1dn\u00e9m p\u0159\u00edpad\u011b nepodobal tomu, co se d\u011bje v p\u0159irozen\u00fdch vaj\u00ed\u010dk\u00e1ch.<\/span><\/p>\n<p><span dir=\"auto\">Kdy\u017e se vaj\u00ed\u010dka tvo\u0159\u00ed p\u0159irozen\u011b, odpov\u00eddaj\u00edc\u00ed chromozomy od matky a otce se sp\u00e1ruj\u00ed, se\u0159ad\u00ed a uspo\u0159\u00e1dan\u011b odd\u011bl\u00ed. Jeden se dostane do vaj\u00ed\u010dka, druh\u00fd je odhozen. To se d\u011bje p\u0159esn\u00fdm a uspo\u0159\u00e1dan\u00fdm zp\u016fsobem. V t\u011bchto rekonstruovan\u00fdch bu\u0148k\u00e1ch v\u0161ak byly chromozomy rozm\u00edst\u011bny n\u00e1hodn\u011b. N\u011bkter\u00e9 bu\u0148ky si zachovaly ob\u011b kopie ur\u010dit\u00fdch chromozom\u016f, zat\u00edmco jin\u00e9 je \u00fapln\u011b ztratily.<\/span><\/p>\n<p><span dir=\"auto\">U zb\u00fdvaj\u00edc\u00edch 46 % embry\u00ed k \u017e\u00e1dn\u00e9mu d\u011blen\u00ed v\u016fbec nedo\u0161lo. V\u011bt\u0161ina si zachovala v\u0161ech 46 chromozom\u016f, m\u00edsto aby se zmen\u0161ila na 23 jako u p\u0159irozen\u00fdch vaj\u00ed\u010dek.<\/span><\/p>\n<p><span dir=\"auto\">U embry\u00ed, jejich\u017e chromozomy se rozd\u011blily, se po\u010dty zna\u010dn\u011b li\u0161ily. N\u011bkter\u00e1 nakonec m\u011bla pouze 3 chromozomy, jin\u00e1 a\u017e 43. V\u011bt\u0161ina se nach\u00e1zela n\u011bkde uprost\u0159ed, kolem 23, ale to, kter\u00e9 konkr\u00e9tn\u00ed chromozomy z\u016fstaly a kter\u00e9 zmizely, bylo v podstat\u011b s\u00e1zkou pro ka\u017ed\u00fd p\u00e1r.<\/span><\/p>\n<p><span dir=\"auto\">Jeden chromozom se choval podivn\u011b. Chromozom 8 konzistentn\u011b pos\u00edlal mat\u010dinu kopii na jedno m\u00edsto a otcovu kopii na jin\u00e9, m\u00edsto aby si vyb\u00edral n\u00e1hodn\u011b. Pro\u010d se to stalo, z\u016fst\u00e1v\u00e1 z\u00e1hadou.<\/span><\/p>\n<h3 class=\"wp-block-heading has-medium-font-size\"><strong><span dir=\"auto\">V\u011bt\u0161ina embry\u00ed se zastavila brzy<\/span><\/strong><\/h3>\n<p><span dir=\"auto\">Kdy\u017e v\u011bdci oplodnili rekonstruovan\u00e9 bu\u0148ky a provedli aktiva\u010dn\u00ed o\u0161et\u0159en\u00ed, v\u011bt\u0161ina z nich se rozd\u011blila do v\u00edce bun\u011bk. Pouze 8,8 % se v\u0161ak vyvinulo do blastocyst, co\u017e je st\u00e1dium, ve kter\u00e9m se embrya obvykle p\u0159en\u00e1\u0161ej\u00ed b\u011bhem IVF l\u00e9\u010dby. Pro srovn\u00e1n\u00ed, tohoto st\u00e1dia dos\u00e1hlo 59 % norm\u00e1ln\u011b oplodn\u011bn\u00fdch kontroln\u00edch vaj\u00ed\u010dek.<\/span><\/p>\n<p><span dir=\"auto\">Anal\u00fdza jednotliv\u00fdch bun\u011bk odhalila r\u016fzn\u00e9 slo\u017een\u00ed chromozom\u016f. N\u011bkter\u00e1 embrya byla jednotn\u00e1, p\u0159i\u010dem\u017e v\u0161echny bu\u0148ky obsahovaly stejnou sm\u011bs chromozom\u016f spermi\u00ed a ko\u017en\u00edch bun\u011bk. Jin\u00e1 byla mozaikov\u00e1, p\u0159i\u010dem\u017e r\u016fzn\u00e9 bu\u0148ky m\u011bly r\u016fzn\u00e9 kombinace chromozom\u016f. T\u00e9m\u011b\u0159 v\u0161echna embrya obsahovala plnou sadu 23 chromozom\u016f spermi\u00ed, a\u010dkoli jednomu mozaikov\u00e9mu embryu n\u011bkter\u00e9 chromozomy spermi\u00ed chyb\u011bly. Po\u010det a p\u016fvod chromozom\u016f ko\u017en\u00edch bun\u011bk se zna\u010dn\u011b li\u0161ily, co\u017e vedlo k chromozom\u00e1ln\u00ed nerovnov\u00e1ze, kter\u00e1 pravd\u011bpodobn\u011b vysv\u011btluje, pro\u010d se v\u011bt\u0161ina embry\u00ed zastavila ve v\u00fdvoji.<\/span><\/p>\n<p><span dir=\"auto\">V\u011bdci nekultivovali embrya d\u00e9le ne\u017e \u0161est\u00fd den, \u010d\u00edm\u017e dodr\u017eovali etick\u00e9 z\u00e1sady pro v\u00fdzkum s lidsk\u00fdmi embryi.<\/span><\/p>\n<h3 class=\"wp-block-heading has-medium-font-size\"><strong><span dir=\"auto\">Roky pr\u00e1ce p\u0159ed klinick\u00fdm pou\u017eit\u00edm<\/span><\/strong><\/h3>\n<p><span dir=\"auto\">V\u011bdci zd\u016fraz\u0148uj\u00ed, \u017ee se jedn\u00e1 pouze o ran\u00fd laboratorn\u00ed experiment a nikoli o l\u00e9\u010dbu. Ne\u017e tohle n\u011bkomu pom\u016f\u017ee, mus\u00ed b\u00fdt vy\u0159e\u0161eno n\u011bkolik d\u016fle\u017eit\u00fdch biologick\u00fdch probl\u00e9m\u016f.<\/span><\/p>\n<p><span dir=\"auto\">Zaprv\u00e9, n\u00e1hodn\u00e9 t\u0159\u00edd\u011bn\u00ed chromozom\u016f nem\u00e1 nic spole\u010dn\u00e9ho s p\u0159irozenou tvorbou vaj\u00ed\u010dek, p\u0159i kter\u00e9 se chromozomy p\u00e1ruj\u00ed a odd\u011bluj\u00ed organizovan\u00fdm zp\u016fsobem. Zadruh\u00e9, p\u0159irozen\u00e9 vaj\u00ed\u010dka p\u0159esouvaj\u00ed genetick\u00fd materi\u00e1l mezi p\u00e1ry chromozom\u016f procesem zvan\u00fdm rekombinace, \u010d\u00edm\u017e vytv\u00e1\u0159ej\u00ed genetickou rozmanitost. V tomto p\u0159\u00edpad\u011b tomu tak v\u016fbec nebylo. Zat\u0159et\u00ed, v\u00fdsledn\u00e1 chromozom\u00e1ln\u00ed nerovnov\u00e1ha by t\u00e9m\u011b\u0159 jist\u011b zabr\u00e1nila \u00fasp\u011b\u0161n\u00e9mu t\u011bhotenstv\u00ed.<\/span><\/p>\n<p><span dir=\"auto\">Vedouc\u00ed v\u00fdzkumn\u00edk Shoukhrat Mitalipov poznamenal, \u017ee t\u00fdm zat\u00edm nem\u016f\u017ee \u0159\u00edci, zda embrya selhala kv\u016fli nespr\u00e1vn\u00e9mu po\u010dtu chromozom\u016f, nebo kv\u016fli nespr\u00e1vn\u011b p\u0159eprogramovan\u00e9 DNA ko\u017en\u00edch bun\u011bk, kter\u00e1 se nechov\u00e1 jako DNA vaj\u00ed\u010dka. Pravd\u011bpodobn\u011b k tomu p\u0159isp\u011bly oba faktory.<\/span><\/p>\n<p><span dir=\"auto\">Podobn\u00e9 experimenty na my\u0161\u00edch vedly k \u017eiv\u00fdm potomk\u016fm, ale reprodukce my\u0161\u00ed a lid\u00ed se natolik li\u0161\u00ed, \u017ee \u00fasp\u011bch u my\u0161\u00ed nen\u00ed z\u00e1rukou, \u017ee tento p\u0159\u00edstup lze extrapolovat na \u010dlov\u011bka.<\/span><\/p>\n<p><span dir=\"auto\">Pokud v\u011bdci nakonec dok\u00e1\u017eou tyto probl\u00e9my vy\u0159e\u0161it, mohl by tento typ technologie pomoci \u017een\u00e1m, kter\u00e9 se narodily bez vaje\u010dn\u00edk\u016f nebo jejich\u017e vaj\u00ed\u010dka byla po\u0161kozena l\u00e9\u010dbou rakoviny, nemoc\u00ed nebo st\u00e1rnut\u00edm. V sou\u010dasn\u00e9 dob\u011b mohou tyto pacientky pou\u017e\u00edvat pouze darovan\u00e1 vaj\u00ed\u010dka nebo zv\u00e1\u017eit adopci.<\/span><\/p>\n<p><span dir=\"auto\">Pozoruhodn\u00fd fakt: P\u0159irozen\u00fd v\u00fdvoj vaj\u00ed\u010dek u lid\u00ed trv\u00e1 v\u00edce ne\u017e deset let, za\u010d\u00edn\u00e1 p\u0159ed narozen\u00edm a kon\u010d\u00ed a\u017e po pubert\u011b. Laboratorn\u00ed p\u0159\u00edstup by mohl tento \u010dasov\u00fd r\u00e1mec zkr\u00e1tit na t\u00fddny, a\u010dkoli d\u016fle\u017eit\u00e9 v\u011bdeck\u00e9 ot\u00e1zky z\u016fst\u00e1vaj\u00ed nezodpov\u011bzeny.<\/span><\/p>\n<p><span dir=\"auto\">V\u00fdzkum byl financov\u00e1n organizacemi Open Philanthropy, Haploid Gamete Research Foundation, Longevity Impetus a Oregon Health &amp; Science University.
