Nalezli m\u011b\u0159iteln\u00e9 mno\u017estv\u00ed zbytkov\u00e9 plazmidov\u00e9 DNA \u2013 a v lahvi\u010dk\u00e1ch Pfizer i dal\u0161\u00ed fragment obsahuj\u00edc\u00ed tzv.\u00a0<\/span><strong><span dir=\"auto\">sekvence promotoru a zesilova\u010de SV40<\/span><\/strong><span dir=\"auto\">\u00a0.<\/span><\/p>\n<p><span dir=\"auto\">Kl\u00ed\u010dov\u00e1 ot\u00e1zka: P\u0159ekra\u010duj\u00ed tyto zbytky DNA mezin\u00e1rodn\u011b platn\u00e9 limity \u2013 a pokud ano, jak\u00fd to m\u00e1 v\u00fdznam pro bezpe\u010dnost a schv\u00e1len\u00ed?<\/span><\/p>\n<h3 class=\"wp-block-heading\"><span dir=\"auto\">N\u00e1vrh a metody studie<\/span><\/h3>\n<ul>\n<li><strong><span dir=\"auto\">Vzorky:<\/span><\/strong><span dir=\"auto\">\u00a032 lahvi\u010dek ze 16 \u0161ar\u017e\u00ed (10 Pfizer \/ 22 Moderna) z kanadsk\u00fdch l\u00e9k\u00e1ren.<\/span><\/li>\n<li><strong><span dir=\"auto\">Testy:<\/span><\/strong><\/li>\n<\/ul>\n<ul>\n<li style=\"list-style-type: none;\">\n<ul class=\"wp-block-list\">\n<li><span dir=\"auto\">qPCR pro spike sekvenci, plasmid ori a elementy SV40.<\/span><\/li>\n<li><span dir=\"auto\">Fluorometrie pro m\u011b\u0159en\u00ed celkov\u00e9ho mno\u017estv\u00ed nukleov\u00e9 kyseliny (v\u010detn\u011b DNA).<\/span><\/li>\n<li><span dir=\"auto\">Nanoporov\u00e9 sekvenov\u00e1n\u00ed lahvi\u010dky pro odhad d\u00e9lky fragmentu.<\/span><\/li>\n<li><span dir=\"auto\">Test DN\u00e1zy I k ur\u010den\u00ed, zda byla DNA chr\u00e1n\u011bna v lipidov\u00fdch nano\u010d\u00e1stic\u00edch (LNP).<\/span><\/li>\n<\/ul>\n<\/li>\n<li><strong><span dir=\"auto\">Dopln\u011bk:<\/span><\/strong><span dir=\"auto\">\u00a0Explorativn\u00ed korelace hladin DNA s hl\u00e1\u0161en\u00edmi ne\u017e\u00e1douc\u00edch \u00fa\u010dink\u016f VAERS.<\/span><\/li>\n<\/ul>\n<h3 class=\"wp-block-heading\"><span dir=\"auto\">Kl\u00ed\u010dov\u00e9 v\u00fdsledky<\/span><\/h3>\n<ul>\n<li><strong><span dir=\"auto\">qPCR:<\/span><\/strong><span dir=\"auto\">\u00a0V\u0161echny lahvi\u010dky Pfizer byly pozitivn\u00ed na Spike, Ori a SV40; lahvi\u010dky Moderna byly pozitivn\u00ed na Spike\/Ori, ale negativn\u00ed na SV40.<\/span><\/li>\n<li><strong><span dir=\"auto\">Mno\u017estv\u00ed:<\/span><\/strong><span dir=\"auto\">\u00a0Pfizer: 0,22\u20137,28 ng DNA\/d\u00e1vka; Moderna: 0,01\u20130,78 ng. Ve dvou \u0161ar\u017e\u00edch p\u0159\u00edpravku Pfizer p\u0159ekro\u010dily hladiny SV40 limit 10 ng stanoven\u00fd WHO\/FDA.<\/span><\/li>\n<li><strong><span dir=\"auto\">Fluorometrie:<\/span><\/strong><span dir=\"auto\">\u00a0V\u00fdrazn\u011b vy\u0161\u0161\u00ed hodnoty \u2013 Pfizer: 371\u20131 548 ng\/d\u00e1vka, Moderna: 1 130\u20136 280 ng\/d\u00e1vka. Tyto hodnoty jsou 36\u2013627kr\u00e1t vy\u0161\u0161\u00ed ne\u017e doporu\u010den\u00ed WHO\/FDA.<\/span><\/li>\n<li><strong><span dir=\"auto\">Nanop\u00f3r:<\/span><\/strong><span dir=\"auto\">\u00a0Fragmenty o pr\u016fm\u011brn\u00e9 d\u00e9lce 214 bp a\u017e 3,5 kb.<\/span><\/li>\n<li><strong><span dir=\"auto\">DN\u00e1za I:<\/span><\/strong><span dir=\"auto\">\u00a0DNA z\u016fstala p\u0159ev\u00e1\u017en\u011b stabiln\u00ed \u2013 co\u017e nazna\u010duje ochranu LNP.<\/span><\/li>\n<li><strong><span dir=\"auto\">Extrapolace:<\/span><\/strong><span dir=\"auto\">\u00a0Miliardy fragment\u016f DNA na d\u00e1vku, pravd\u011bpodobn\u011b propustn\u00e9 pro bu\u0148ky prost\u0159ednictv\u00edm zapouzd\u0159en\u00ed LNP.<\/span><\/li>\n<\/ul>\n<h3 class=\"wp-block-heading\"><span dir=\"auto\">Omezen\u00ed<\/span><\/h3>\n<ul>\n<li><strong><span dir=\"auto\">P\u016fvod vzorku:<\/span><\/strong><span dir=\"auto\">\u00a0lahvi\u010dky z l\u00e9k\u00e1ren, \u017e\u00e1dn\u00fd v\u00fdb\u011br v\u00fdrobce, sekvenov\u00e1na pouze jedna lahvi\u010dka.<\/span><\/li>\n<li><strong><span dir=\"auto\">Metodologie:<\/span><\/strong><span dir=\"auto\">\u00a0qPCR a fluorometrie nejsou validov\u00e1ny podle mezin\u00e1rodn\u00edch standard\u016f; mo\u017en\u00e9 zkreslen\u00ed m\u011b\u0159en\u00ed v d\u016fsledku zbytk\u016f RNA nebo LNP.<\/span><\/li>\n<li><strong><span dir=\"auto\">Rozd\u00edly:<\/span><\/strong><span dir=\"auto\">\u00a0qPCR a fluorometrie poskytuj\u00ed velmi odli\u0161n\u00e9 v\u00fdsledky.<\/span><\/li>\n<li><strong><span dir=\"auto\">Limity:<\/span><\/strong><span dir=\"auto\">\u00a0Hodnota WHO\/FDA (10 ng\/d\u00e1vka) poch\u00e1z\u00ed ze star\u0161\u00edch typ\u016f vakc\u00edn; jej\u00ed p\u0159enositelnost na mRNA LNP je nejasn\u00e1.<\/span><\/li>\n<li><strong><span dir=\"auto\">Klinick\u00fd v\u00fdznam:<\/span><\/strong><span dir=\"auto\">\u00a0Nejsou k dispozici \u017e\u00e1dn\u00e9 \u00fadaje o zdravotn\u00edch v\u00fdsledc\u00edch. Srovn\u00e1n\u00ed VAERS jsou \u010dist\u011b pr\u016fzkumn\u00e1 a nejsou pr\u016fkazn\u00e1.<\/span><\/li>\n<li><strong><span dir=\"auto\">St\u0159et z\u00e1jm\u016f:<\/span><\/strong><span dir=\"auto\">\u00a0Jeden z autor\u016f pracuje pro v\u00fdrobce pou\u017eit\u00fdch testovac\u00edch sad.<\/span><\/li>\n<\/ul>\n<h3 class=\"wp-block-heading\"><span dir=\"auto\">Z\u00e1v\u011br a v\u00fdznam<\/span><\/h3>\n<p><span dir=\"auto\">Studie poukazuje na\u00a0<\/span><strong><span dir=\"auto\">analyticky v\u011brohodn\u00fd sign\u00e1l<\/span><\/strong><span dir=\"auto\">\u00a0: zbytkovou DNA, n\u011bkdy ve vysok\u00e9m mno\u017estv\u00ed, a tak\u00e9 sekvence SV40 v lahvi\u010dk\u00e1ch Pfizeru. To neznamen\u00e1\u00a0<\/span><strong><span dir=\"auto\">,<\/span><\/strong><span dir=\"auto\">\u00a0\u017ee jsou viry SV40 p\u0159\u00edtomny nebo \u017ee automaticky existuje zdravotn\u00ed riziko.<\/span><\/p>\n<p><span dir=\"auto\">Zji\u0161t\u011bn\u00ed v\u0161ak vyvol\u00e1vaj\u00ed ot\u00e1zky pro\u00a0<\/span><strong><span dir=\"auto\">v\u00fdrobce a regula\u010dn\u00ed org\u00e1ny<\/span><\/strong><span dir=\"auto\">\u00a0. Zejm\u00e9na kolik DNA (a jak\u00fd typ) je p\u0159ijateln\u00e9 ve vakc\u00edn\u00e1ch s mRNA LNP a zda je t\u0159eba upravit st\u00e1vaj\u00edc\u00ed limity.<\/span><\/p>\n<p><span dir=\"auto\">Auto\u0159i po\u017eaduj\u00ed nez\u00e1visl\u00e9, standardizovan\u00e9 n\u00e1sledn\u00e9 studie s v\u011bt\u0161\u00edm vzorkem.
