T\u00fdm tedy rychle do\u0161el k z\u00e1v\u011bru, \u017ee b\u00edl\u00e9 vl\u00e1knit\u00e9 sra\u017eeniny NEJSOU jen norm\u00e1ln\u00ed sra\u017eeniny krve.<\/p>\n<p>Naopak v\u0161echny t\u0159i anal\u00fdzy ICP-MS uk\u00e1zaly, \u017ee b\u00edl\u00e9 vl\u00e1knit\u00e9 sra\u017eeniny obsahuj\u00ed velmi vysok\u00e1 mno\u017estv\u00ed fosforu, s\u00edry a v n\u011bkter\u00fdch p\u0159\u00edpadech i c\u00ednu.<\/p>\n<p>Nap\u0159\u00edklad koncentrace fosforu v norm\u00e1ln\u00ed krvi je mezi 100 a\u017e 1 000 ppm. Av\u0161ak ve vzorc\u00edch b\u00edl\u00e9 vl\u00e1knit\u00e9 sra\u017eeniny bylo nam\u011b\u0159eno mno\u017estv\u00ed fosforu a\u017e 5 000 ppm! Jak uvid\u00edte, toto vysok\u00e9 mno\u017estv\u00ed fosforu se v tomto p\u0159\u00edb\u011bhu st\u00e1v\u00e1 velmi d\u016fle\u017eit\u00fdm.<\/p>\n<p>Hlavn\u00ed v\u011bdec Greg Harrison uva\u017eoval, zda toto abnorm\u00e1ln\u00ed vysok\u00e9 mno\u017estv\u00ed fosforu v b\u00edl\u00fdch vl\u00e1knit\u00fdch sra\u017eenin\u00e1ch m\u016f\u017ee b\u00fdt \u010d\u00e1ste\u010dn\u011b nebo z velk\u00e9 \u010d\u00e1sti zodpov\u011bdn\u00e9 za to, pro\u010d se sra\u017eeniny tvo\u0159\u00ed. Probl\u00e9m byl vy\u0159e\u0161en sn\u00ed\u017een\u00edm s\u00edly pod\u00e1van\u00e9ho heparinu. Dnes, pokud pacienti vykazuj\u00ed n\u011bjak\u00e9 probl\u00e9my s heparinem, mohou p\u0159ej\u00edt na jin\u00e1 antikoagulancia jako Eliquis, Pradaxa nebo Xarelto.<\/p>\n<p>Greg se domn\u00edval, \u017ee pokud k tomuto jevu m\u016f\u017ee doj\u00edt z p\u0159\u00edli\u0161 velk\u00e9ho mno\u017estv\u00ed s\u00edry v krvi, m\u016f\u017ee se to st\u00e1t i tehdy, je-li v krvi p\u0159\u00edli\u0161 mnoho fosforu.<\/p>\n<p>To jej p\u0159ivedlo k jeho sou\u010dasn\u00e9 \u201eteorii fosforylace\u201c fosfolipidov\u00fdch nano\u010d\u00e1stic z vakc\u00edny a hrotov\u00e9ho proteinu (bu\u010f z viru nebo vakc\u00edny), kter\u00e9 se v\u00e1\u017e\u00ed s p\u0159irozen\u00fdm fibrinogenem v t\u011ble za vzniku nep\u0159irozen\u00e9ho, \u200b\u200bzkroucen\u00e9ho, \u200b\u200btj. nespr\u00e1vn\u011b poskl\u00e1dan\u00e9ho poly.<\/p>\n<p>Takovou sra\u017eeninu v\u0161ak p\u0159irozen\u00fd enzym na\u0161eho t\u011bla, plazmin, nedok\u00e1\u017ee rozpustit.<\/p>\n<p>Greg a jeho t\u00fdm z\u00edskali potvrzen\u00ed, \u017ee s p\u0159irozen\u00fdm fibrinogenem v t\u011ble se skute\u010dn\u011b d\u011bje n\u011bco zvl\u00e1\u0161tn\u00edho pot\u00e9, co dostali v\u00fdsledky sv\u00e9ho dal\u0161\u00edho testu. Na n\u011bkolika vzorc\u00edch b\u00edl\u00fdch vl\u00e1knit\u00fdch sra\u017eenin byla provedena anal\u00fdza vysoce v\u00fdkonnou kapalinovou chromatografi\u00ed (HPLC).<\/p>\n<p>Tento test identifikuje a m\u011b\u0159\u00ed mno\u017estv\u00ed r\u016fzn\u00fdch \u201eslou\u010denin\u201c ve vzorku. Nen\u00ed p\u0159ekvapen\u00edm, \u017ee anal\u00fdza HPLC zjistila, \u017ee asi 56% b\u00edl\u00e9 vl\u00e1knit\u00e9 sra\u017eeniny tvo\u0159\u00ed t\u011blu p\u0159irozen\u00fd fibrinogen. Pom\u011br subkomponent\u016f fibrinogenu byl v\u0161ak drasticky mimo normu!<\/p>\n<p>Kdy\u017e se fibrinogen v na\u0161\u00ed krevn\u00ed plazm\u011b jako sou\u010d\u00e1st norm\u00e1ln\u00edho procesu sr\u00e1\u017een\u00ed p\u0159em\u011bn\u00ed z tekut\u00e9ho stavu na b\u00edlou pevnou l\u00e1tku zvanou \u201efibrin\u201c, t\u0159i podslo\u017eky fibrinogenu se spoj\u00ed a vytvo\u0159\u00ed vl\u00e1kno fibrinu.<\/p>\n<p>Tyto \u0159et\u011bzce fibrinogenu zvan\u00e9 alfa, beta a gama se propl\u00e9taj\u00ed v pom\u011bru 1:1:1 a vytv\u00e1\u0159ej\u00ed norm\u00e1ln\u00ed vl\u00e1kno fibrinu.<\/p>\n<p>Anal\u00fdza HPLC v\u0161ak odhalila, \u017ee 36% b\u00edl\u00e9 vl\u00e1knit\u00e9 sra\u017eeniny tvo\u0159\u00ed beta \u0159et\u011bzec fibrinogenu, 16% sra\u017eeniny tvo\u0159\u00ed gama \u0159et\u011bzec fibrinogenu a pouze 4% sra\u017eeniny tvo\u0159\u00ed alfa \u0159et\u011bzec fibrinogenu. To p\u0159edstavuje bizarn\u00ed pom\u011br 1:9:4 na rozd\u00edl od norm\u00e1ln\u00edho pom\u011bru 1:1:1.<\/p>\n<p>Na z\u00e1klad\u011b toho t\u00fdm dosp\u011bl k z\u00e1v\u011bru, \u017ee n\u011bjak\u00e1 velmi neobvykl\u00e1 chemick\u00e1 reakce v t\u011ble jist\u011b m\u011bn\u00ed proces, kter\u00fdm se tekut\u00fd fibrinogen p\u0159em\u011b\u0148uje na norm\u00e1ln\u00ed pevn\u00fd fibrin.<\/p>\n<p>Dal\u0161\u00edm vod\u00edtkem bylo, \u017ee trombin, kter\u00fd p\u016fsob\u00ed jako katalyz\u00e1tor p\u0159i norm\u00e1ln\u00ed p\u0159em\u011bn\u011b fibrinogenu na fibrin, nebyl v\u016fbec p\u0159\u00edtomen v b\u00edl\u00fdch vl\u00e1knit\u00fdch sra\u017eenin\u00e1ch. Reakci tedy vyvolalo n\u011bco jin\u00e9ho.<\/p>\n<p>Nejen\u017ee se zm\u011bnila p\u0159em\u011bna fibrinogenu na fibrin, ale Greg m\u011bl podez\u0159en\u00ed, \u017ee fibrinogen se v procesu \u201enespr\u00e1vn\u011b poskl\u00e1d\u00e1\u201c v d\u016fsledku jeho interakce s hrotov\u00fdm proteinem a vakc\u00ednov\u00fdmi fosfolipidov\u00fdmi nano\u010d\u00e1sticemi.<\/p>\n<p>Greg se proto rozhodl otestovat tyto nespr\u00e1vn\u011b slo\u017een\u00e9 proteiny, zn\u00e1m\u00e9 tak\u00e9 jako \u201eamyloidn\u00ed proteiny\u201c, pomoc\u00ed n\u011bkolika r\u016fzn\u00fdch test\u016f. Barven\u00ed barvou \u201eKongo Red\u201c se nejprve uskute\u010dnilo na n\u011bkolika vzorc\u00edch b\u00edl\u00fdch vl\u00e1knit\u00fdch sra\u017eenin a vr\u00e1tilo pozitivn\u00ed v\u00fdsledek na p\u0159\u00edtomnost amyloidu.<\/p>\n<p>Aby se d\u00e1le potvrdila p\u0159\u00edtomnost amyloidn\u00edch protein\u016f v b\u00edl\u00fdch vl\u00e1knit\u00fdch sra\u017eenin\u00e1ch, Greg\u016fv t\u00fdm tak\u00e9 provedl testov\u00e1n\u00ed Thioflavin-T na n\u011bkolika vzorc\u00edch sra\u017eenin, co\u017e je dal\u0161\u00ed technika barven\u00ed, p\u0159i kter\u00e9 se vzorky pod ultrafialov\u00fdm sv\u011btlem rozsv\u00edt\u00ed na \u201ezeleno\u201c v p\u0159\u00edpad\u011b, kdy se zjist\u00ed p\u0159\u00edtomnost amy.<\/p>\n<p>No a v\u00fdsledek byl, \u017ee v\u0161echny vzorky b\u00edl\u00e9 vl\u00e1knit\u00e9 sra\u017eeniny se rozsv\u00edtily zelen\u011b. Ve skute\u010dnosti se pozd\u011bji zjistilo, \u017ee i bez zabarven\u00ed vzorku b\u00edl\u00e9 vl\u00e1knit\u00e9 sra\u017eeniny ST\u00c1LE sv\u00edtily pod ultrafialov\u00fdm sv\u011btlem zelen\u011b, co\u017e potvrdilo silnou p\u0159\u00edtomnost amyloidu!<\/p>\n<p>Potvrzen\u00ed amyloidn\u00edch protein\u016f v b\u00edl\u00fdch vl\u00e1knit\u00fdch sra\u017eenin\u00e1ch nebylo pro Grega a jeho t\u00fdm \u00fapln\u00fdm p\u0159ekvapen\u00edm. V\u0161echny t\u0159i vzorky sra\u017eenin byly testov\u00e1ny jako \u201epozitivn\u00ed\u201c!<\/p>\n<h2>Tak\u017ee, co to v\u0161echno znamen\u00e1 pro sv\u011bt?<\/h2>\n<p>Pokud jde o amyloidn\u00ed sra\u017eeniny, tedy mikrosra\u017eeniny, kter\u00e9 blokuj\u00ed tepny a mal\u00e9 krevn\u00ed c\u00e9vy, ty mohou zp\u016fsobit tichou hypoxii (nedokysli\u010den\u00ed) nebo org\u00e1novou dysfunkci nap\u0159\u00edklad v mozku, ledvin\u00e1ch nebo srdci, ani\u017e by byla zji\u0161t\u011bna zjevn\u00e1 tromb\u00f3za.<\/p>\n<p>Pokud jde o b\u00edl\u00e9 vl\u00e1knit\u00e9 sra\u017eeniny nebo makrozra\u017eeniny, tyto mohou zp\u016fsobit zjevn\u00e9 mozkov\u00e9 mrtvice a srde\u010dn\u00ed infarkty.<\/p>\n<p>Pokud jde o prionov\u00e1 onemocn\u011bn\u00ed, mohou se objevit neurovaskul\u00e1rn\u00ed nebo neurodegenerativn\u00ed poruchy, a to mnohem rychleji ne\u017e za desetilet\u00ed \u2013 co\u017e je \u010das, kolik obvykle trv\u00e1, ne\u017e se tyto poruchy projev\u00ed.<\/p>\n<p>Nap\u0159\u00edklad u mlad\u0161\u00edch lid\u00ed se mohou objevit neurovaskul\u00e1rn\u00ed poruchy, jako jsou m\u00edrn\u00e1 kognitivn\u00ed porucha, mozkov\u00e1 mlha, t\u0159es a zm\u011bny chov\u00e1n\u00ed.
