Pomoc\u00ed fluorometru Qubit v\u011bdci kvantifikovali pr\u016fm\u011brn\u011b 216 ng zbytkov\u00e9 DNA na d\u00e1vku, co\u017e se zv\u00fd\u0161ilo a\u017e na 5 160 ng na d\u00e1vku po o\u0161et\u0159en\u00ed Triton-X-100, l\u00e1tkou uvol\u0148uj\u00edc\u00ed DNA. Sekvenov\u00e1n\u00ed nov\u00e9 generace potvrdilo kompletn\u00ed sekvenci plazmidov\u00e9 DNA vakc\u00edny s 98,3% pokryt\u00edm a vysokou hloubkou sekvenov\u00e1n\u00ed. V\u00fdsledky nazna\u010duj\u00ed ne\u010distoty DNA ve vakc\u00edn\u011b a vyvol\u00e1vaj\u00ed ot\u00e1zky ohledn\u011b mo\u017en\u00fdch rizik integrace DNA b\u011bhem pod\u00e1v\u00e1n\u00ed.<\/p>\n<p>Raoult zd\u016fraznil pot\u0159ebu dal\u0161\u00edch studi\u00ed, proto\u017ee tato studie zkoumala pouze jednu lahvi\u010dku a nezjistila \u017e\u00e1dn\u00e9 klinick\u00e9 \u00fa\u010dinky kontaminant\u016f DNA. V p\u0159edtisku popisuje mo\u017en\u00e1 rizika:<\/p>\n<p>\u201eVysok\u00e1 mno\u017estv\u00ed plazmidov\u00e9 DNA na d\u00e1vku vakc\u00edny by mohla p\u0159edstavovat potenci\u00e1ln\u00ed riziko pro integraci do lidsk\u00e9ho genomu, zejm\u00e9na prost\u0159ednictv\u00edm balen\u00ed do kationtov\u00fdch lipid\u016f. Podobn\u00e9 mechanismy byly pozorov\u00e1ny u genov\u00fdch terapi\u00ed na b\u00e1zi DNA, kde bylo 10\u201320 % bun\u011bk transfekov\u00e1no a 1\u201310 % bylo stabiln\u011b integrov\u00e1no.\u201c P\u0159ipou\u0161t\u00ed v\u0161ak, \u017ee riziko genomick\u00e9 integrace a tedy mo\u017en\u00e9ho spu\u0161t\u011bn\u00ed onkogen\u016f nebo deaktivace Supresory n\u00e1dor\u016f jsou velmi n\u00edzk\u00e9.<\/p>\n<p>Studie m\u00e1 v\u0161ak omezen\u00ed: zam\u011b\u0159uje se na jednu lahvi\u010dku, d\u00edky \u010demu\u017e by v\u00fdsledky mohly b\u00fdt nereprezentativn\u00ed, a neposkytuje p\u0159\u00edm\u00e9 d\u016fkazy o zdravotn\u00edch rizic\u00edch. Zji\u0161t\u011bn\u00e1 mno\u017estv\u00ed DNA by teoreticky mohla p\u0159isp\u011bt k bun\u011b\u010dn\u00e9 integraci, ale to z\u016fst\u00e1v\u00e1 spekulativn\u00ed.<\/p>\n<p>Celkov\u011b pr\u00e1ce zd\u016fraz\u0148uje pot\u0159ebu p\u0159\u00edsn\u00fdch kontrol kvality p\u0159i v\u00fdrob\u011b vakc\u00edn a vyz\u00fdv\u00e1 k dal\u0161\u00edmu v\u00fdzkumu bezpe\u010dnosti a potenci\u00e1ln\u00edho dopadu kontaminace DNA.
