Expert na genomiku Kevin McKernan kritizoval metody studie, ale tvrdil, \u017ee kontaminace je st\u00e1le nad povolen\u00fdmi limity a \u017ee sou\u010dasn\u00e9 p\u0159edpisy jsou &#8222;zcela nedostate\u010dn\u00e9.&#8220;<\/span><\/strong><\/em><\/p>\n<p><span>Nov\u00e1 recenzovan\u00e1 studie vyvol\u00e1v\u00e1 obavy ohledn\u011b metod pou\u017e\u00edvan\u00fdch k testov\u00e1n\u00ed mRNA vakc\u00edny Comirnaty COVID-19 vyroben\u00e9 spole\u010dnostmi Pfizer a BioNTech na mo\u017enou kontaminaci DNA.<\/span><\/p>\n<p><span>Ve studii zve\u0159ejn\u011bn\u00e9 tento m\u011bs\u00edc v Methods and Protocols n\u011bme\u010dt\u00ed v\u011bdci Brigitte K\u00f6nig a J\u00fcrgen O. Kirchner zpochyb\u0148uj\u00ed spolehlivost techniky kvantitativn\u00ed PCR (qPCR), kterou spole\u010dnost Pfizer-BioNTech pou\u017e\u00edv\u00e1 k m\u011b\u0159en\u00ed kontaminace DNA v \u00fa\u010dinn\u00e9 l\u00e1tce pou\u017eit\u00e9 ve vakc\u00edn\u011b. .<\/span><\/p>\n<p><span>V\u011bdci experimentovali s rozpou\u0161t\u011bn\u00edm Comirnatyho lipidov\u00fdch nano\u010d\u00e1stic. Na\u0161li ne\u010distoty DNA, kter\u00e9 byly 360 a\u017e 534kr\u00e1t vy\u0161\u0161\u00ed ne\u017e limit 10 ng (nanogram\u016f) na d\u00e1vku stanoven\u00fd regul\u00e1tory po cel\u00e9m sv\u011bt\u011b.<\/span><\/p>\n<p><span>V\u00fdzkumn\u00edci navrhli, \u017ee metody fluorescen\u010dn\u00ed spektroskopie by mohly spolehliv\u011bji kvantifikovat celkov\u00e9 mno\u017estv\u00ed kontaminace DNA v kone\u010dn\u00e9m vakc\u00ednov\u00e9m produktu p\u0159ipraven\u00e9m k pou\u017eit\u00ed.<\/span><\/p>\n<p><span>Kevin McKernan, hlavn\u00ed v\u011bdeck\u00fd pracovn\u00edk a zakladatel Medicinal Genomics, \u0159ekl The Defender, \u017ee a\u010dkoli auto\u0159i upozornili na n\u011bkter\u00e9 d\u016fle\u017eit\u00e9 body t\u00fdkaj\u00edc\u00ed se kontaminace DNA v mRNA vakc\u00edn\u00e1ch COVID-19, fluorometrick\u00e1 barviva mohou b\u00fdt nespolehliv\u00e1, co\u017e vede k nadm\u011brn\u00fdm nam\u011b\u0159en\u00fdm hodnot\u00e1m.<\/span><\/p>\n<p class=\"has-medium-font-size\"><strong><span>&#8222;Masivn\u00ed nedostate\u010dn\u00e9 hl\u00e1\u0161en\u00ed kontaminace DNA&#8220;<\/span><\/strong><\/p>\n<p><span>V\u00fdrobci jako Pfizer-BioNTech pou\u017e\u00edvaj\u00ed testy kontaminace DNA zalo\u017een\u00e9 na metod\u011b qPCR aplikovan\u00e9 na \u00fa\u010dinnou l\u00e1tku vakc\u00edny p\u0159ed jej\u00edm zkombinov\u00e1n\u00edm s lipidov\u00fdmi nano\u010d\u00e1sticemi.<\/span><\/p>\n<p><span>K\u00f6nig a Kirchner pouk\u00e1zali na to, \u017ee test qPCR zkoum\u00e1 pouze mal\u00fd segment o 69 p\u00e1rech b\u00e1z\u00ed p\u016fvodn\u00edho templ\u00e1tu DNA o 7824 p\u00e1rech b\u00e1z\u00ed pou\u017eit\u00e9ho k v\u00fdrob\u011b vakc\u00edny mRNA.<\/span><\/p>\n<p><span>To znamen\u00e1, \u017ee spole\u010dnost Pfizer zkontroluje m\u00e9n\u011b ne\u017e 1 % p\u016fvodn\u00edho p\u0159\u00edsp\u011bvku. Zb\u00fdvaj\u00edc\u00edch 99 % z\u016fst\u00e1v\u00e1 neanalyzov\u00e1no, co\u017e vede k \u201emasivn\u00edmu podhodnocen\u00ed kontaminace DNA,\u201c uvedli v\u011bdci.<\/span><\/p>\n<p><span>V\u011bdci tak\u00e9 tvrdili, \u017ee tento mal\u00fd segment m\u016f\u017ee b\u00fdt zni\u010den b\u011bhem procesu enzymov\u00e9ho \u0161t\u011bpen\u00ed jinou rychlost\u00ed ne\u017e zb\u00fdvaj\u00edc\u00ed fragmenty templ\u00e1tu DNA, co\u017e d\u00e1le komplikuje p\u0159esn\u00e1 m\u011b\u0159en\u00ed.<\/span><\/p>\n<p><span>Dal\u0161\u00edm komplikuj\u00edc\u00edm faktorem je, \u017ee c\u00edlov\u00e1 sekvence qPCR se p\u0159ekr\u00fdv\u00e1 s \u00fasekem DNA naz\u00fdvan\u00fdm promotor T7, kter\u00fd se pou\u017e\u00edv\u00e1 k produkci mRNA. Bun\u011b\u010dn\u00e9 mechanismy nebo vedlej\u0161\u00ed produkty by se mohly v\u00e1zat na tuto promotorovou oblast a blokovat tak detekci testem qPCR.<\/span><\/p>\n<p><span>David Speicher, Ph.D., spoluautor s McKernanem a dal\u0161\u00edmi p\u0159edtiskov\u00e9 studie o fragmentech DNA ve vakc\u00edn\u00e1ch Moderna a Pfizer COVID-19, vyj\u00e1d\u0159il podobn\u00e9 obavy.<\/span><\/p>\n<p><span>PCR m\u016f\u017ee kvantifikovat pouze specifickou sekvenci DNA\/RNA, na kterou c\u00edl\u00ed pou\u017eit\u00e9 primery, vysv\u011btlil The Defender. Pokud se v t\u00e9to c\u00edlov\u00e9 sekvenci vyskytnou zlomy nebo mutace, DNA nebude replikov\u00e1na a z\u00e1t\u011b\u017e bude podhodnocena.\u201c<\/span><\/p>\n<p><span>&#8222;P\u0159edpokl\u00e1d\u00e1 se tak\u00e9, \u017ee DNA ve vakc\u00edn\u011b poch\u00e1z\u00ed pouze z plazmidu a ne z bakteri\u00e1ln\u00edho nebo jin\u00e9ho zdroje,&#8220; \u0159ekl Speicher.<\/span><\/p>\n<p><span>McKernan pouk\u00e1zal na dal\u0161\u00ed probl\u00e9m: Regula\u010dn\u00ed org\u00e1ny umo\u017e\u0148uj\u00ed spole\u010dnosti Pfizer pou\u017e\u00edvat qPCR k m\u011b\u0159en\u00ed DNA a fluorometrii k m\u011b\u0159en\u00ed RNA.<\/span><\/p>\n<p><span>\u201eP\u0159edpisy EMA [Evropsk\u00e9 l\u00e9kov\u00e9 agentury] vy\u017eaduj\u00ed pom\u011brov\u00e9 m\u011b\u0159en\u00ed RNA:DNA,\u201c \u0159ekl. &#8222;Pom\u011bry by se nem\u011bly m\u011b\u0159it v centimetrech pro RNA a metrech pro DNA.&#8220;<\/span><\/p>\n<p><span>\u0158ekl, \u017ee Pfizer by m\u011bl m\u011b\u0159it RNA i DNA pomoc\u00ed fluorometrie nebo qPCR. &#8222;Kdy\u017e si dovol\u00ed kombinovat n\u00e1stroje, jako jsou tyto, umo\u017e\u0148uj\u00ed p\u0159\u00edm\u00fd podvod.&#8220;<\/span><\/p>\n<p><span>McKernan tak\u00e9 sd\u00edlel \u010d\u00e1st patentov\u00e9 p\u0159ihl\u00e1\u0161ky Moderny, kter\u00e1 uzn\u00e1v\u00e1, \u017ee qPCR je nevhodn\u00e1 pro m\u011b\u0159en\u00ed mal\u00fdch fragment\u016f DNA.<\/span><\/p>\n<p class=\"has-medium-font-size\"><span style=\"font-size: 18pt;\"><strong>&#8222;U\u017e nediskutujeme o tom, zda jsou nahr\u00e1vky kontaminovan\u00e9&#8220;<\/strong><\/span><\/p>\n<p><span>Aby se p\u0159ede\u0161lo n\u00e1strah\u00e1m qPCR, kter\u00e1 detekuje pouze malou \u010d\u00e1st kontaminovan\u00e9 DNA, K\u00f6nig a Kirchner navrhli pou\u017e\u00edt techniky fluorescen\u010dn\u00ed spektroskopie, jako je Qubit, ke kvantifikaci celkov\u00e9ho obsahu DNA v kone\u010dn\u00e9m vakc\u00ednov\u00e9m produktu.<\/span><\/p>\n<p><span>Tyto metody vyu\u017e\u00edvaj\u00ed fluorescen\u010dn\u00ed barviva, kter\u00e1 se specificky v\u00e1\u017eou na nukleov\u00e9 kyseliny, jako je DNA a RNA.<\/span><\/p>\n<p><span>V jejich experimentech fluorescen\u010dn\u00ed technikou s Comirnaty byla po rozbit\u00ed nano\u010d\u00e1stic zji\u0161t\u011bna kontaminace DNA, kter\u00e1 byla v\u00fdrazn\u011b nad limitem 10 ng\/d\u00e1vku.<\/span><\/p>\n<div class=\"wp-block-image\" style=\"text-align: center;\">\n<figure class=\"aligncenter size-large\"><img decoding=\"async\" class=\"aligncenter\" src=\"https:\/\/i0.wp.com\/childrenshealthdefense.org\/wp-content\/uploads\/1-Konig-and-Kirchner-figure-2.jpg?w=800&amp;ssl=1\" alt=\"\" data-recalc-dims=\"1\" \/><figcaption class=\"wp-element-caption\"><em><span>Obr. 2. RN\u00e1za je enzym, kter\u00fd odstra\u0148uje RNA, tak\u017ee p\u0159i m\u011b\u0159en\u00ed DNA nedoch\u00e1z\u00ed k interferenci s RNA.<\/span><\/p>\n<p><span>Bez t\u00e9to kontroly Koenig a Kirchner \u201evytvo\u0159ili snadn\u00fd c\u00edl pro sv\u00e9 kritiky,\u201c \u0159ekl.<\/span><\/p>\n<p><span>Ve v\u00fdzkumu p\u0159ipravovan\u00e9m k publikaci McKernan uvedl, \u017ee n\u011bkolik laborato\u0159\u00ed, kter\u00e9 prov\u00e1d\u011bj\u00ed experimenty s RN\u00e1zou, zaznamenalo 10n\u00e1sobn\u00e9 sn\u00ed\u017een\u00ed sign\u00e1lu DNA ve fluorometrii.<\/span><\/p>\n<p><span>&#8222;To znamen\u00e1, \u017ee kontaminace DNA je st\u00e1le v\u00fdrazn\u011b nad limitem FDA [US Food and Drug Administration],&#8220; \u0159\u00edk\u00e1 McKernan.
