\u201eP\u0159eklasifikace benzimidazolov\u00fdch l\u00e9k\u016f proti parazit\u016fm jako selektivn\u00edch protirakovinn\u00fdch chemoterapeutik (2023)\u201c v\u00fdzkumn\u00edci zjistili, \u017ee fenbendazol m\u00e1 dva prim\u00e1rn\u00ed mechanismy \u00fa\u010dinku proti rakovinn\u00fdm bu\u0148k\u00e1m.<\/p>\n<p>Za prv\u00e9, l\u00e9k vykazuje antimitotickou aktivitu, co\u017e znamen\u00e1, \u017ee potla\u010duje polymeraci tubulinu vazbou na tubul\u00ednov\u00e1 m\u00edsta rychle se d\u011bl\u00edc\u00edch bun\u011bk.\u00a0Tento \u00fa\u010dinek zp\u016fsobuje zastaven\u00ed bun\u011b\u010dn\u00e9ho cyklu.<\/p>\n<p>Za druh\u00e9, l\u00e9k vyvol\u00e1v\u00e1 oxida\u010dn\u00ed stres, kter\u00fd naru\u0161uje bun\u011b\u010dn\u00e9 metabolick\u00e9 procesy.\u00a0Zastavuje vychyt\u00e1v\u00e1n\u00ed gluk\u00f3zy (hlavn\u00ed potravy rakovinov\u00fdch bun\u011bk), potla\u010duje kl\u00ed\u010dov\u00e9 enzymy a sni\u017euje \u00farove\u0148 dostupn\u00e9 energie ATP, co\u017e vede k apopt\u00f3ze, tedy smrti jak parazit\u016f, tak rakovinov\u00fdch bun\u011bk.<\/p>\n<p>V\u011bdci ale zjistili, \u017ee benzimidazolov\u00e9 l\u00e9ky maj\u00ed n\u00edzkou rozpustnost ve vod\u011b, co\u017e zt\u011b\u017euje jejich pou\u017eit\u00ed v klinick\u00fdch aplikac\u00edch.\u00a0To vedlo v\u00fdzkumn\u00edky ke kombinaci l\u00e9ku s nano\u010d\u00e1sticemi pro efektivn\u011bj\u0161\u00ed aplikace in vivo (v \u017eiv\u00fdch organismech).<\/p>\n<p>V sou\u010dasn\u00e9 dob\u011b neexistuj\u00ed \u017e\u00e1dn\u00e9 klinick\u00e9 zkou\u0161ky pro fenbendazol, ale existuj\u00ed klinick\u00e9 zkou\u0161ky pro podobn\u00fd l\u00e9k v t\u00e9to t\u0159\u00edd\u011b, naz\u00fdvan\u00fd mebendazol (pou\u017e\u00edvan\u00fd nap\u0159\u00edklad p\u0159i hl\u00edstech u lid\u00ed).\u00a0Tento l\u00e9k se ukazuje jako slibn\u00e1 adjuvantn\u00ed (podp\u016frn\u00e1) l\u00e9\u010dba rakoviny tlust\u00e9ho st\u0159eva a mozkov\u00fdch n\u00e1dor\u016f v kombinaci s jin\u00fdmi antiprotozo\u00e1ln\u00edmi l\u00e1tkami.<\/p>\n<p>Existuj\u00ed tak\u00e9 d\u016fkazy, \u017ee benzimidazolov\u00e9 l\u00e9ky zcitlivuj\u00ed n\u00e1dorov\u00e9 bu\u0148ky, tak\u017ee se mohou st\u00e1t n\u00e1chyln\u011bj\u0161\u00edmi k\u00a0<a href=\"https:\/\/www.badatel.net\/co-siri-rakovinu-rychlejsie-chemoterapia-ozarovanie-alebo-vakciny-proti-covidu\/\">radia\u010dn\u00ed terapii<\/a>\u00a0.<\/p>\n<p>Ve studii \u201eProtirakovinov\u00fd \u00fa\u010dinek nano\u010d\u00e1stic PLGA se zabudovan\u00fdm fenbendazolem p\u0159i rakovin\u011b vaje\u010dn\u00edk\u016f\u201c v\u00fdzkumn\u00edci zjistili, \u017ee protirakovinn\u00e9 \u00fa\u010dinky fenbendazolu se neprojevovaly na my\u0161\u00edch modelech, dokud nebyl p\u0159\u00edpravek p\u0159ipraven s pou\u017eit\u00edm nano\u010d\u00e1stic k jeho \u00fa\u010dinn\u00e9mu dod\u00e1v\u00e1n\u00ed bu\u0148k\u00e1m.<\/p>\n<p>Nano\u010d\u00e1stice zvy\u0161uj\u00ed biologickou dostupnost l\u00e9\u010diva, co\u017e mu umo\u017e\u0148uje \u00fa\u010dinn\u011b interferovat s polymerac\u00ed mikrotubul\u016f.<\/p>\n<p>Proto\u017ee l\u00e9k blokoval progresi bun\u011b\u010dn\u00e9ho cyklu, zv\u00fd\u0161il stabilitu proteinu p53 a vyvolal apopt\u00f3zu rakovinov\u00fdch bun\u011bk.\u00a0Nakonec l\u00e9k sn\u00ed\u017eil bun\u011b\u010dnou proliferaci (mno\u017een\u00ed) chemosenzitivn\u00edch i chemorezistentn\u00edch \u00fa\u010dinn\u00fd proti rakovin\u011b prsu a chemorezistentn\u00ed rakovin\u011b tlust\u00e9ho st\u0159eva a kone\u010dn\u00edku<\/h2>\n<p>Ve studii \u201eRedoxem zprost\u0159edkovan\u00e1 protirakovinn\u00e1 aktivita antiparazitick\u00e9ho l\u00e9ku fenbendazolu v trojn\u00e1sobn\u011b negativn\u00edch bu\u0148k\u00e1ch rakoviny prsu\u201c v\u00fdzkumn\u00edci testovali fenbendazol na trojn\u00e1sobn\u011b negativn\u00edch bu\u0148k\u00e1ch rakoviny prsu, v\u010detn\u011b vysoce metastatick\u00e9ho typu.<\/p>\n<p>L\u00e9\u010divo vyvolalo v rakovinov\u00fdch bu\u0148k\u00e1ch oxida\u010dn\u00ed stres, \u010d\u00edm\u017e se staly zraniteln\u011bj\u0161\u00edmi v\u016f\u010di apopt\u00f3ze (bun\u011b\u010dn\u00e9 smrti).<\/p>\n<p>V dal\u0161\u00ed studii z roku 2023 byl p\u0159\u00edpravek testov\u00e1n na my\u0161\u00edch lymfomech a bu\u0148k\u00e1ch sleziny, aby se zjistilo, zda existuje rozd\u00edl mezi zab\u00edjen\u00edm rakovinov\u00fdch bun\u011bk a zachov\u00e1n\u00edm zdrav\u00fdch bun\u011bk sleziny.<\/p>\n<p>V\u00fdzkum uk\u00e1zal, \u017ee fenbendazol vytv\u00e1\u0159\u00ed reaktivn\u00ed formy kysl\u00edku a zp\u016fsobuje bun\u011b\u010dnou smrt bun\u011bk lymfomu, ale nepo\u0161kozuje norm\u00e1ln\u00ed bu\u0148ky sleziny.\u00a0Uk\u00e1zalo se tak\u00e9, \u017ee l\u00e9k je m\u00e9n\u011b toxick\u00fd pro zdrav\u00e9 bu\u0148ky ne\u017e tradi\u010dn\u00ed chemoterapeutick\u00e1 \u010dinidla, ale je podobn\u011b smrteln\u00fd pro rakovinov\u00e9 bu\u0148ky.<\/p>\n<p>Ve studii z roku 2022 byl fenbendazol testov\u00e1n proti chemorezistentn\u00edm bu\u0148k\u00e1m kolorekt\u00e1ln\u00edho karcinomu.\u00a0V\u011bdci zjistili, \u017ee fenbendazol zde m\u00e1 \u0161est mechanism\u016f \u00fa\u010dinku.<\/p>\n<p>L\u00e9\u010divo spou\u0161t\u00ed apopt\u00f3zu bez ovlivn\u011bn\u00ed exprese genu pro protirakovinn\u00fd protein p53.\u00a0Tak\u00e9 indukuje apopt\u00f3zu Beclin-1 a zp\u016fsobuje zastaven\u00ed bun\u011b\u010dn\u00e9ho cyklu rakovinov\u00fdch bun\u011bk ve f\u00e1zi G2\/M.<\/p>\n<p>Krom\u011b toho l\u00e9k zp\u016fsobuje autofagii (o\u010distu bun\u011bk), nekropt\u00f3zu (z\u00e1n\u011btlivou bun\u011b\u010dnou smrt) a feropt\u00f3zu (bun\u011b\u010dnou smrt z\u00e1vislou na \u017eeleze), p\u0159i\u010dem\u017e zab\u00edj\u00ed i ty nejodoln\u011bj\u0161\u00ed rakovinov\u00e9 bu\u0148ky.<\/p>\n<h2>Fenbendazol by mohl b\u00fdt p\u0159eklasifikov\u00e1n k l\u00e9\u010db\u011b lidsk\u00e9ho glioblastomu a leuk\u00e9mie<\/h2>\n<p>Dal\u0161\u00ed studie z roku 2022 poskytuje d\u016fkaz, \u017ee benzimidazoly indukuj\u00ed apopt\u00f3zu a pyropt\u00f3zu lidsk\u00fdch rakovinn\u00fdch bun\u011bk glioblastomu (GBM).\u00a0V\u011bdci zjistili, \u017ee fenbendazol potla\u010duje synt\u00e9zu DNA v z\u00e1vislosti na d\u00e1vce.<\/p>\n<p>Krom\u011b toho l\u00e9k inhiboval migraci bun\u011bk, \u010d\u00edm\u017e zabr\u00e1nil dal\u0161\u00edmu metast\u00e1zov\u00e1n\u00ed bun\u011bk glioblastomu.\u00a0V\u00fdzkumn\u00edci podrobn\u011b popsali p\u0159\u00edmou cestu, kterou l\u00e9k zp\u016fsobil programovanou bun\u011b\u010dnou smrt in vivo (v re\u00e1ln\u00fdch podm\u00ednk\u00e1ch \u017eiv\u00e9ho organismu, nikoli ve zkumavce) a kde p\u0159esn\u011b zp\u016fsobil zastaven\u00ed bun\u011b\u010dn\u00e9ho cyklu.<\/p>\n<p>Nakonec dv\u011b studie z roku 2020 zjistily, \u017ee benzimidazoly by se mohly p\u0159em\u011bnit na protin\u00e1dorov\u00e9 l\u00e9ky pro lidi a mohly by se \u00fasp\u011b\u0161n\u011b pou\u017e\u00edt i proti leuk\u00e9mii.<\/p>\n<p>V prvn\u00ed studii v\u00fdzkumn\u00edci chv\u00e1lili benzimidazoly za to, \u017ee jsou dob\u0159e tolerov\u00e1ny u zv\u00ed\u0159at iu lid\u00ed, s velmi mal\u00fdmi vedlej\u0161\u00edmi \u00fa\u010dinky.\u00a0L\u00e9ky se nejen v\u00e1\u017e\u00ed na \u03b2-tubulin parazitick\u00fdch \u010derv\u016f, co\u017e zp\u016fsobuje jejich imobilizaci a smrt, ale maj\u00ed tak\u00e9 protirakovinn\u00e9 aktivity, jako jsou:<\/p>\n<ul>\n<li>naru\u0161en\u00ed polymerace mikrotubul\u016f<\/li>\n<li>inhibice \u017eivotaschopnosti rakovinov\u00fdch bun\u011bk<\/li>\n<li>migrace, invaze a indukce apopt\u00f3zy<\/li>\n<li>autofagie rakovinov\u00e9 bu\u0148ky<\/li>\n<\/ul>\n<p>V dal\u0161\u00ed studii z roku 2020 bylo zji\u0161t\u011bno, \u017ee fenbendazol m\u00e1 specificky protirakovinnou aktivitu v bu\u0148k\u00e1ch HL-60, kter\u00e9 jsou bun\u011b\u010dnou lini\u00ed lidsk\u00e9 leuk\u00e9mie.\u00a0V z\u00e1vislosti na koncentraci l\u00e9\u010divo sni\u017euje metabolickou aktivitu a mitochondri\u00e1ln\u00ed membr\u00e1nov\u00fd potenci\u00e1l leukemick\u00fdch bun\u011bk.\u00a0Nakonec l\u00e9k zv\u00fd\u0161il apopt\u00f3zu a nekr\u00f3zu leukemick\u00fdch bun\u011bk.<\/p>\n<p>Po v\u0161em slibn\u00e9m v\u00fdzkumu tohoto antiparazitick\u00e9ho l\u00e9ku by nem\u011bl b\u00fdt d\u016fvod ke zdr\u017eov\u00e1n\u00ed klinick\u00fdch zkou\u0161ek fenbendazolu p\u0159i l\u00e9\u010db\u011b rakoviny u lid\u00ed, aby se mohl
