Tento syntetick\u00fd estrogen se pou\u017e\u00edval k profylaxi potratu, p\u0159ed\u010dasn\u00e9ho porodu a nitrod\u011blo\u017en\u00edho odum\u0159en\u00ed plodu, [1] ale i jako nouzov\u00e1 antikoncepce. [2]<\/p>\n<p>P\u0159edpokl\u00e1d\u00e1 se, \u017ee celosv\u011btov\u011b bylo v obdob\u00ed let 1940-1971 p\u0159ibli\u017en\u011b 6 milion\u016f matek a jejich plod\u016f vystaven\u00fdch vlivu dietylstilboestrolu (d\u00e1le DES). [3]<\/p>\n<p>Jak se uk\u00e1zalo pozd\u011bji, DES, kter\u00fd se pova\u017eoval za z\u00e1zra\u010dn\u00fd l\u00e9k (miraculous drug), [4] p\u016fsobil jako tzv. trans-placent\u00e1rn\u00ed karcinogen, proch\u00e1zel p\u0159es placentu matek a zp\u016fsoboval jejich dosp\u00edvaj\u00edc\u00edm dcer\u00e1m (p\u0159ibli\u017en\u011b ve v\u011bku 7-19 let) rakovinu reproduk\u010dn\u00edch org\u00e1n\u016f, zejm\u00e9na adenokarcinom pochvy nebo d\u011blo\u017en\u00edho \u010d\u00edpku. [5] [6] [7] [8] Byly zaznamen\u00e1ny i jin\u00e9 typy rakoviny u dcer t\u011bchto matek, nap\u0159\u00edklad smrteln\u00fd teratom vaje\u010dn\u00edku u 12 let\u00e9ho d\u011bv\u010d\u00e1tka, [9] nebo adenom hypof\u00fdzy u adolescentky ve v\u011bku 18 let. [10]<\/p>\n<p>Krom\u011b ji\u017e zm\u00edn\u011bn\u00fdch onkologick\u00fdch onemocn\u011bn\u00ed, dcery, jejich\u017e matk\u00e1m byl pod\u00e1v\u00e1n DES b\u011bhem t\u011bhotenstv\u00ed, m\u011bly v\u00fdznamn\u011b naru\u0161eny reproduk\u010dn\u00ed zdrav\u00ed projevuj\u00edc\u00ed se v\u00fdrazn\u011b vy\u0161\u0161\u00edm v\u00fdskytem zm\u011bn na d\u011blo\u017en\u00edm \u010d\u00edpku a pochv\u011b, [11] neplodnost\u00ed, spont\u00e1nn\u00edmi potraty, p\u0159ed\u010dasn\u00fdmi porody, mimod\u011blo\u017en\u00edm t\u011bhotenstv\u00edm, porody mrtv\u00e9ho d\u00edt\u011bte, p\u0159ed\u010dasnou menopauzou, ale i rakovinou prsu . [27]<\/p>\n<p>Uveden\u00e1 zji\u0161t\u011bn\u00ed potvrdila, \u017ee DES p\u016fsob\u00ed negativn\u011b na zdrav\u00ed potomk\u016f nejen prost\u0159ednictv\u00edm genotoxick\u00fdch \u00fa\u010dink\u016f, [28] ale zejm\u00e9na prost\u0159ednictv\u00edm epigenetick\u00fdch \u00fa\u010dink\u016f, kter\u00e9 se p\u0159en\u00e1\u0161ej\u00ed na dal\u0161\u00ed generace, tedy i na ty, kter\u00e9 nebyly p\u0159\u00edmo vystaveny jeho \u00fa\u010dink\u016fm (tzv. transgenera\u010dn\u00ed \u00fa\u010dinky). [29] [30] Epigenetick\u00e9 mechanismy, kter\u00e9 jsou velmi citliv\u00e9 v\u016f\u010di extern\u00edm intervenc\u00edm, nem\u011bn\u00ed sekvenci DNA, ale reguluj\u00ed genovou expresi na transkrip\u010dn\u00ed a posttranskrip\u010dn\u00ed \u00farovni. Klasick\u00e9 epigenetick\u00e9 mechanismy zahrnuj\u00ed metylaci DNA, modifikace histon\u016f a regulaci mikroRNA (miRNA). Podobn\u011b jako dietylstilbestrol i mRNA injekce proti SARS-CoV-2 se vehementn\u011b doporu\u010duje nejzraniteln\u011bj\u0161\u00edm populac\u00edm jako jsou t\u011bhotn\u00e9 \u017eeny [32] a dosp\u00edvaj\u00edc\u00ed d\u011bti. [33]<\/p>\n<p>Obdob\u00ed embryon\u00e1ln\u00edho v\u00fdvoje jako\u017e i obdob\u00ed dosp\u00edv\u00e1n\u00ed (adolescence) je charakteristick\u00e9 prudk\u00fdm r\u016fstem a mno\u017een\u00edm bun\u011bk, a proto vy\u017eaduje mimo\u0159\u00e1dnou pozornost. Puberta startuje f\u00e1zi rychl\u00e9ho r\u016fstu a zr\u00e1n\u00ed zejm\u00e9na reproduk\u010dn\u00edho, svalov\u011b kostern\u00edho, nervov\u00e9ho, endokrinn\u00edho, metabolick\u00e9ho, imunitn\u00edho a kardio-vaskul\u00e1rn\u00edho syst\u00e9mu. Z tohoto d\u016fvodu je dosp\u00edv\u00e1n\u00ed zvl\u00e1\u0161\u0165 citlivou f\u00e1z\u00ed lidsk\u00e9ho \u017eivota b\u011bhem n\u00ed\u017e mohou negativn\u00ed vlivy v\u00fdznamn\u011b ovlivnit nejen v\u00fdvin a zdrav\u00ed jedince ale i zdrav\u00ed p\u0159\u00ed\u0161t\u00ed generace. [34]<\/p>\n<p>V souvislosti s o\u010dkov\u00e1n\u00edm dosp\u00edvaj\u00edc\u00edch d\u011bt\u00ed injekcemi proti SARS-CoV-2 je d\u016fle\u017eit\u00e9 upozornit na tzv. hypotalamickou kontrolu puberty. Je zn\u00e1mo, \u017ee p\u0159ibli\u017en\u011b 10% hypotalamick\u00fdch gen\u016f (zn\u00e1m\u00fdch pod n\u00e1zvem Geny Souvisej\u00edc\u00ed s n\u00e1dorem -Tumor Related Genes, TRG) zvy\u0161uj\u00edc\u00edch svou aktivitu b\u011bhem puberty z\u00e1rove\u0148 kontroluje tvorbu nebo potla\u010dov\u00e1n\u00ed n\u00e1dor\u016f.<\/p>\n<p>Roth a kol. identifikovali 11 gen\u016f souvisej\u00edc\u00edch s TRG s\u00edt\u00ed z nich\u017e 6 bylo identifikov\u00e1no jako geny potla\u010duj\u00edc\u00ed n\u00e1dory (nap\u0159. p53 , SASH1 , CUTL1 ) a 5 jako geny zp\u016fsobuj\u00edc\u00ed n\u00e1dory (nap\u0159. CCND1 a AF1Q , jejich\u017e naru\u0161en\u00e1 exprese byla pozorov\u00e1na nap\u0159. p\u0159i rakovin\u011b prsu [35 ] [36] ). [37] Pokud mi je zn\u00e1mo, nejsou k dispozici studie, kter\u00e9 by zkoumaly vliv injekc\u00ed proti SARS-CoV-2 na TRG hypotalamickou s\u00ed\u0165, kter\u00e1 ve spolupr\u00e1ci s dal\u0161\u00edmi geny hraje v\u00fdznamnou roli nejen ve startu puberty a regulaci reproduk\u010dn\u00edho v\u00fdvinu, ale tak\u00e9 v tvorb\u011b nebo potla\u010dov\u00e1n\u00ed n\u00e1dor\u016f. [38] [39]<\/p>\n<p>Velmi d\u016fle\u017eitou roli v tomto obdob\u00ed hraj\u00ed epigenetick\u00e9 mechanismy, kter\u00e9 aktivuj\u00ed nebo deaktivuj\u00ed \u010d\u00e1sti genomu v konkr\u00e9tn\u00edm \u010dase a na konkr\u00e9tn\u00edm m\u00edst\u011b. [40] Pokud mi je zn\u00e1mo, neexistuj\u00ed epidemiologick\u00e9 \u00fadaje o interakci injekc\u00ed proti SARS-CoV-2 s epigenetick\u00fdmi mechanismy.<\/p>\n<p>2.
