\u017ee by mohly slou\u017eit jako \u201e\u00fa\u010dinn\u00e9 n\u00e1hra\u017eky vakc\u00edn\u201c.<\/li>\n<li>Protil\u00e1tky &#8212; TAU-1109 a TAU-2310 &#8212; se v\u00e1\u017eou na jinou oblast spike proteinu ne\u017e jin\u00e9 protil\u00e1tky, oblast, kter\u00e1 nem\u00e1 mnoho mutac\u00ed.<\/li>\n<li>TAU-1109 je 92% \u00fa\u010dinn\u00fd p\u0159i neutralizaci omikronov\u00e9ho kmene a 90% \u00fa\u010dinn\u00fd p\u0159i neutralizaci delta kmene<\/li>\n<li>TAU-2310 m\u00e1 84% \u00fa\u010dinnost p\u0159i neutralizaci omikronov\u00e9ho kmene a 97% \u00fa\u010dinnost proti delta variant\u011b<\/li>\n<li>V\u011bdci se domn\u00edvaj\u00ed, \u017ee p\u0159i \u00fa\u010dinn\u00e9 l\u00e9\u010db\u011b protil\u00e1tkami \u201enemus\u00edme d\u00e1vat posilovac\u00ed d\u00e1vky cel\u00e9 populaci poka\u017ed\u00e9, kdy\u017e se objev\u00ed nov\u00e1 varianta\u201c.<\/li>\n<\/ul>\n<p>V\u011bdci z Tel Avivsk\u00e9 univerzity objevili dv\u011b protil\u00e1tky, kter\u00e9 neutralizuj\u00ed SARS-CoV-2 \u2013 a v\u0161echny jeho varianty \u2013 tak \u00fa\u010dinn\u011b, \u017ee v\u011b\u0159\u00ed, \u017ee by protil\u00e1tky mohly slou\u017eit jako \u201e\u00fa\u010dinn\u00e9 n\u00e1hra\u017eky vakc\u00edny\u201c.\u00a0Jednou z do o\u010d\u00ed bij\u00edc\u00edch chyb vakc\u00edn proti COVID-19 je jejich nedostate\u010dn\u00e1 \u00fa\u010dinnost proti vznikaj\u00edc\u00edm kmen\u016fm COVID-19.<\/p>\n<p>P\u0159i v\u00fdb\u011bru spike proteinu jako z\u00e1kladu pro vakc\u00ednu COVID-19 zvolili v\u011bdci protein, o kter\u00e9m je nejen zn\u00e1mo, \u017ee je toxick\u00fd pro \u010dlov\u011bka, ale tak\u00e9 \u017ee nen\u00ed sou\u010d\u00e1st\u00ed viru, kter\u00fd vyvol\u00e1v\u00e1 nejlep\u0161\u00ed imunitn\u00ed odpov\u011b\u010f.<\/p>\n<p>\u0160pi\u010dkov\u00fd protein rychle mutuje a prakticky neguje jakoukoli ochranu, kterou nab\u00edz\u00ed o\u010dkov\u00e1n\u00ed kr\u00e1tce po pod\u00e1n\u00ed.\u00a0V\u00fdsledkem je zd\u00e1nliv\u011b nekone\u010dn\u00e1 s\u00e9rie ka\u017edoro\u010dn\u00edch o\u010dkov\u00e1n\u00ed a p\u0159eo\u010dkov\u00e1n\u00ed, kter\u00e9 mohou poskytnout pouze rychle se sni\u017euj\u00edc\u00ed ochranu.\u00a0Pokud se potvrd\u00ed zji\u0161t\u011bn\u00ed izraelsk\u00fdch v\u011bdc\u016f a protil\u00e1tky budou tak \u00fa\u010dinn\u00e9, jak se p\u0159edpokl\u00e1d\u00e1, p\u0159eo\u010dkov\u00e1n\u00ed COVID-19 by mohlo b\u00fdt \u00fapln\u011b vynech\u00e1no.<\/p>\n<p class=\"has-medium-font-size\"><span style=\"font-size: 18pt;\"><strong>P\u0159edchoz\u00ed l\u00e9\u010dba protil\u00e1tkami m\u011bla sm\u00ed\u0161en\u00e9 v\u00fdsledky<\/strong><\/span><\/p>\n<p>Americk\u00fd \u00fa\u0159ad pro potraviny a l\u00e9\u010diva (FDA) schv\u00e1lil n\u011bkolik koktejl\u016f monoklon\u00e1ln\u00edch protil\u00e1tek pro l\u00e9\u010dbu COVID-19.\u00a0\u00da\u010dinnost r\u016fzn\u00fdch variant se v\u0161ak li\u0161ila, n\u011bkter\u00e9 se staly ne\u00fa\u010dinn\u00fdmi a jin\u00e9 z\u016fstaly \u00fa\u010dinn\u00e9.<\/p>\n<p>&#8222;To nazna\u010duje, \u017ee n\u011bkter\u00e9 protil\u00e1tky vyvolan\u00e9 infekc\u00ed jsou citliv\u011bj\u0161\u00ed na variace ne\u017e jin\u00e9, a \u017ee je t\u0159eba vz\u00edt v \u00favahu \u0161\u00ed\u0159i specifi\u010dnosti protil\u00e1tek a nejen jejich \u00fa\u010dinnost,&#8220; p\u00ed\u0161\u00ed v\u011bdci z Clinical Division Microbiology and Immunology at the Sacklerova l\u00e9ka\u0159sk\u00e1 fakulta, Tel Aviv University v \u010dasopise Communications Biology.<\/p>\n<p>Nap\u0159\u00edklad v lednu 2022 FDA omezila pou\u017eit\u00ed dvou l\u00e9\u010debn\u00fdch monoklon\u00e1ln\u00edch protil\u00e1tek \u2013 bamlanivimab a etesevimab, kter\u00e9 se pod\u00e1vaj\u00ed spole\u010dn\u011b, a REGEN-COV (casirivimab a imdevimab) \u2013 na pacienty infikovan\u00e9 variantou, o kter\u00e9 je zn\u00e1mo, \u017ee je infikovan\u00e1. jim.\u00a0Dv\u011b zm\u00edn\u011bn\u00e9 protil\u00e1tkov\u00e9 l\u00e9\u010dby ztratily velkou \u010d\u00e1st sv\u00e9 \u00fa\u010dinnosti proti variant\u011b Omicron, co\u017e vedlo k omezen\u00ed pou\u017eit\u00ed u lid\u00ed infikovan\u00fdch p\u0159\u00edpravkem Omicron.<\/p>\n<p>Na druhou stranu v \u00fanoru 2022 FDA ud\u011blila povolen\u00ed k nouzov\u00e9mu pou\u017eit\u00ed (EUA) pro l\u00e9\u010dbu monoklon\u00e1ln\u00edmi protil\u00e1tkami nazvanou bebtelovimab, kter\u00e1 z\u016fst\u00e1v\u00e1 \u00fa\u010dinn\u00e1 proti omikronov\u00e9 variant\u011b.\u00a0Podle FDA:<\/p>\n<p><em>Bebtelovimab p\u016fsob\u00ed tak, \u017ee se v\u00e1\u017ee na spike protein viru, kter\u00fd zp\u016fsobuje COVID-19, podobn\u011b jako jin\u00e9 monoklon\u00e1ln\u00ed protil\u00e1tky schv\u00e1len\u00e9 pro l\u00e9\u010dbu vysoce rizikov\u00fdch pacient\u016f s m\u00edrn\u00fdm a\u017e st\u0159edn\u011b z\u00e1va\u017en\u00fdm onemocn\u011bn\u00edm COVID-19, u kter\u00fdch bylo prok\u00e1z\u00e1no, \u017ee sni\u017euj\u00ed riziko hospitalizace nebo \u00famrt\u00ed.<\/em><\/p>\n<p>To, jak se protil\u00e1tky v\u00e1\u017eou na spike protein, by mohlo b\u00fdt kl\u00ed\u010dem k jejich kone\u010dn\u00e9 \u00fa\u010dinnosti proti r\u016fzn\u00fdm kmen\u016fm.\u00a0V p\u0159edchoz\u00ed studii proveden\u00e9 v \u0159\u00edjnu 2020 hlavn\u00ed autorka studie Natalia Freund a jej\u00ed kolegov\u00e9 izolovali dev\u011bt protil\u00e1tek od lid\u00ed, kte\u0159\u00ed se zotavili z p\u016fvodn\u00edho kmene COVID-19 v Izraeli.\u00a0Freund vysv\u011btlil v tiskov\u00e9 zpr\u00e1v\u011b:<\/p>\n<p><em>V p\u0159edchoz\u00ed studii jsme uk\u00e1zali, \u017ee r\u016fzn\u00e9 protil\u00e1tky produkovan\u00e9 v reakci na infekci p\u016fvodn\u00edm virem jsou nam\u00ed\u0159eny proti r\u016fzn\u00fdm \u010d\u00e1stem viru.\u00a0Nej\u00fa\u010dinn\u011bj\u0161\u00ed protil\u00e1tky byly ty, kter\u00e9 se nav\u00e1zaly na protein \u201espike\u201c viru na stejn\u00e9m m\u00edst\u011b, kde se bodec v\u00e1\u017ee na bun\u011b\u010dn\u00fd receptor ACE2.<\/em><\/p>\n<p><cite>Samoz\u0159ejm\u011b jsme nebyli jedin\u00ed, kdo tyto protil\u00e1tky izoloval, a glob\u00e1ln\u00ed zdravotnick\u00fd syst\u00e9m je hojn\u011b vyu\u017e\u00edval, dokud se neobjevily r\u016fzn\u00e9 varianty koronaviru, kter\u00e9 u\u010dinily v\u011bt\u0161inu t\u011bchto protil\u00e1tek zbyte\u010dnou.<\/cite><\/p>\n<p class=\"has-medium-font-size\"><strong>Dv\u011b protil\u00e1tky neutralizuj\u00ed v\u0161echny kmeny COVID-19<\/strong><\/p>\n<p>Zde prezentovan\u00e1 studie navazuje na studii z \u0159\u00edjna 2020 a ukazuje dv\u011b protil\u00e1tky &#8211; TAU-1109 a TAU-2310 &#8211; kter\u00e9 se v\u00e1\u017eou na jinou oblast spike proteinu &#8211; oblast, kde je m\u00e1lo mutac\u00ed &#8211; a takov\u00e9 jsou schopny neutralizovat v\u0161echny zn\u00e1m\u00e9 kmeny COVID-19.\u00a0Podle p\u0159\u00edtele:<\/p>\n<p><em>V sou\u010dasn\u00e9 studii se n\u00e1m poda\u0159ilo prok\u00e1zat, \u017ee dv\u011b dal\u0161\u00ed protil\u00e1tky, TAU-1109 a TAU-2310, kter\u00e9 v\u00e1\u017eou virov\u00fd spike protein v jin\u00e9 oblasti, ne\u017e kam se v\u011bt\u0161ina protil\u00e1tek d\u0159\u00edve soust\u0159edila (a proto p\u0159i neutralizaci p\u016fvodn\u00edho kmene byly m\u00e9n\u011b \u00fa\u010dinn\u00e9) jsou ve skute\u010dnosti velmi \u00fa\u010dinn\u00e9 p\u0159i neutralizaci delta a omikronov\u00fdch variant.<\/em><\/p>\n<p>Konkr\u00e9tn\u011b zjistili, \u017ee TAU-1109 neutralizoval 92 % omikronov\u00e9ho kmene a 90 % delta kmene.\u00a0TAU-2310 m\u00e1 84% \u00fa\u010dinnost p\u0159i neutralizaci omikronov\u00e9ho kmene a 97% \u00fa\u010dinnost p\u0159i neutralizaci delta varianty.\u00a0Studie byla provedena ve spolupr\u00e1ci s Kalifornskou univerzitou v San Diegu, kam byly tyto dv\u011b protil\u00e1tky odesl\u00e1ny k dal\u0161\u00edmu testov\u00e1n\u00ed proti \u017eiv\u00fdm vir\u016fm v laboratorn\u00edch kultur\u00e1ch.<\/p>\n<p>Protil\u00e1tky byly tak\u00e9 testov\u00e1ny proti pseudovir\u016fm na Bar Ilan University v Galilei.\u00a0&#8222;V\u00fdsledky byly v obou testech stejn\u00e9 a stejn\u011b povzbudiv\u00e9,&#8220; uv\u00e1d\u00ed tiskov\u00e1 zpr\u00e1va.\u00a0Zaj\u00edmav\u00e9 je, \u017ee mutuj\u00edc\u00ed virus mohl hr\u00e1t roli v tom, \u017ee tyto dv\u011b protil\u00e1tky byly tak \u00fa\u010dinn\u00e9.\u00a0p\u0159\u00edtel vysv\u011btlil:<\/p>\n<p><em>Infek\u010dnost viru se zvy\u0161ovala s ka\u017edou variantou, proto\u017ee poka\u017ed\u00e9 zm\u011bnil aminokyselinovou sekvenci \u010d\u00e1sti spike proteinu, kter\u00e1 se v\u00e1\u017ee na ACE2 receptor, \u010d\u00edm\u017e se zvy\u0161uje jeho infek\u010dnost a z\u00e1rove\u0148 se vyh\u00fdb\u00e1 p\u0159irozen\u00fdm protil\u00e1tk\u00e1m produkovan\u00fdm po o\u010dkov\u00e1n\u00ed.<\/em><\/p>\n<p><cite>Naproti tomu protil\u00e1tky TAU-1109 a TAU-2310 se nev\u00e1\u017eou na vazebn\u00e9 m\u00edsto ACE2 receptoru, ale na jinou oblast spike proteinu &#8211; oblast virov\u00e9 spike, kter\u00e1 z n\u011bjak\u00e9ho d\u016fvodu nepodl\u00e9h\u00e1 mnoha mutac\u00edm &#8211; a jsou proto je \u00fa\u010dinn\u00fd p\u0159i neutralizaci v\u00edce variant vir\u016f.\u00a0Tato zji\u0161t\u011bn\u00ed p\u0159i\u0161la, kdy\u017e jsme testovali v\u0161echny d\u0159\u00edve zn\u00e1m\u00e9 kmeny COVID.<\/cite><\/p>\n<p class=\"has-medium-font-size\"><span style=\"font-size: 18pt;\"><strong>COVID boostery nejsou \u0159e\u0161en\u00edm<\/strong><\/span><\/p>\n<p>Freund v\u011b\u0159\u00ed, \u017ee protil\u00e1tky jsou tak \u00fa\u010dinn\u00e9, \u017ee by mohly p\u0159\u00edpadn\u011b nahradit posilovac\u00ed d\u00e1vky COVID-19.\u00a0To je dobr\u00e1 zpr\u00e1va, proto\u017ee ochrana poskytovan\u00e1 o\u010dkov\u00e1n\u00edm proti COVID-19, v\u010detn\u011b p\u0159eo\u010dkov\u00e1n\u00ed, netrv\u00e1 dlouho.<\/p>\n<p>Studie financovan\u00e1 americk\u00fdm centrem pro kontrolu a prevenci nemoc\u00ed zahrnovala data z 10 st\u00e1t\u016f shrom\u00e1\u017ed\u011bn\u00e1 mezi 26. srpnem 2021 a 22. lednem 2022, tedy obdob\u00edm, kdy kolovala jak delta, tak omikronov\u00e1 varianta.13 B\u011bhem dvou m\u011bs\u00edc\u016f od druh\u00e9ho COVID -19 o\u010dkov\u00e1n\u00ed, ochrana p\u0159ed n\u00e1v\u0161t\u011bvami pohotovosti a urgentn\u00ed l\u00e9\u010dba souvisej\u00edc\u00ed s COVID-19 byla 69 %.\u00a0P\u011bt m\u011bs\u00edc\u016f po o\u010dkov\u00e1n\u00ed tato hodnota klesla na 37 %.<\/p>\n<p>N\u00edzk\u00e1 \u00fa\u010dinnost p\u011bt m\u011bs\u00edc\u016f po prvn\u00edm o\u010dkov\u00e1n\u00ed p\u0159im\u011bla \u00fa\u0159ady k doporu\u010den\u00ed p\u0159eo\u010dkov\u00e1n\u00ed \u2013 a t\u0159et\u00ed o\u010dkov\u00e1n\u00ed zv\u00fd\u0161ilo \u00fa\u010dinnost na 87 %.\u00a0Tento n\u00e1r\u016fst byl v\u0161ak kr\u00e1tkodob\u00fd.\u00a0Ochrana p\u0159ed n\u00e1v\u0161t\u011bvami pohotovosti klesla na 66 % b\u011bhem \u010dty\u0159 a\u017e p\u011bti m\u011bs\u00edc\u016f po p\u0159eo\u010dkov\u00e1n\u00ed a pot\u00e9 p\u011bt m\u011bs\u00edc\u016f nebo d\u00e9le po p\u0159eo\u010dkov\u00e1n\u00ed klesla na pouh\u00fdch 31 %.<\/p>\n<p>Sp\u00ed\u0161e ne\u017e p\u0159ipustit por\u00e1\u017eku, zdravotni\u010dt\u00ed \u00fa\u0159edn\u00edci pl\u00e1nuj\u00ed d\u00e1t je\u0161t\u011b v\u00edce d\u00e1vek sv\u00fdch trapn\u011b ne\u00fa\u010dinn\u00fdch posilovac\u00edch d\u00e1vek pomoc\u00ed nov\u00fdch, takzvan\u00fdch \u201eaktualizovan\u00fdch\u201c v\u00fdst\u0159el\u016f.\u00a0Dne 31. srpna 2022 upravil FDA EUA pro vakc\u00edny COVID-19 Moderna a Pfizer tak, aby umo\u017e\u0148ovaly bivalentn\u00ed formulace jako posilovac\u00ed d\u00e1vky alespo\u0148 dva m\u011bs\u00edce po p\u0159edchoz\u00ed p\u0159eo\u010dkov\u00e1n\u00ed nebo z\u00e1va\u017en\u00e9m z\u00e1kladn\u00edm o\u010dkov\u00e1n\u00ed.<\/p>\n<p>\u201eBivalentn\u00ed vakc\u00edny, kter\u00e9 tak\u00e9 ozna\u010dujeme jako \u201aaktualizovan\u00e9 posilovac\u00ed vakc\u00edny\u2018, obsahuj\u00ed dv\u011b slo\u017eky medi\u00e1torov\u00e9 RNA (mRNA) viru SARS-CoV-2, jednu z p\u016fvodn\u00edho kmene SARS-CoV-2 a druhou nesouc\u00ed Linky BA.4 a BA.5 jsou spole\u010dn\u00e9 pro omikronovou variantu SARS-CoV-2,\u201c uvedl FDA.<\/p>\n<p>Krom\u011b vysok\u00e9 mo\u017enosti, \u017ee ochrana proti COVID-19 po p\u0159eo\u010dkov\u00e1n\u00ed rychle zmiz\u00ed, existuje re\u00e1ln\u00e9 riziko z\u00e1va\u017en\u00fdch ne\u017e\u00e1douc\u00edch \u00fa\u010dink\u016f, pokud jsou protil\u00e1tky ve va\u0161em t\u011ble um\u011ble posilov\u00e1ny opakovan\u00fdmi p\u0159eo\u010dkov\u00e1n\u00ed.<\/p>\n<p>To oklame t\u011blo, aby si myslelo, \u017ee je v\u017edy infikov\u00e1no COVID-19, co\u017e je stav, kter\u00fd m\u016f\u017ee v\u00e9st pouze k \u201ez\u00f3n\u011b smrti\u201c a urychluje rozvoj autoimunitn\u00edch onemocn\u011bn\u00ed, jako je Parkinsonova choroba, Kawasakiho choroba a roztrou\u0161en\u00e1 skler\u00f3za, podle technologick\u00e9ho l\u00eddra a COVID. -19 Analytik Marc Girardot nal\u00e9h\u00e1 na to, abyste se dostali z \u201ez\u00f3ny smrti\u201c vakc\u00edny COVID, ne\u017e bude p\u0159\u00edli\u0161 pozd\u011b<\/p>\n<p class=\"has-medium-font-size\"><span style=\"font-size: 18pt;\"><strong>S protil\u00e1tkovou l\u00e9\u010dbou ji\u017e nebudeme muset pou\u017e\u00edvat \u201eposilova\u010de\u201c<\/strong><\/span><\/p>\n<p>Mo\u017en\u00e1 je sv\u011btlo na konci tunelu, proto\u017ee Freund douf\u00e1, \u017ee \u201ezk\u0159\u00ed\u017een\u011b neutraliza\u010dn\u00ed schopnosti protil\u00e1tek p\u0159irozen\u011b produkovan\u00fdch b\u011bhem infekce divok\u00e9ho typu SARS-CoV-2\u201c by mohly poskytnout alternativu k posilovac\u00edm o\u010dkov\u00e1n\u00edm.\u00a0Vysv\u011btlila:<\/p>\n<p><em>Z d\u016fvod\u016f, kter\u00fdm zat\u00edm pln\u011b nerozum\u00edme, mno\u017estv\u00ed protil\u00e1tek proti COVID-19 po t\u0159ech m\u011bs\u00edc\u00edch v\u00fdrazn\u011b kles\u00e1, proto se lid\u00e9 mohou nakazit i po t\u0159ech o\u010dkov\u00e1n\u00edch.<\/em><\/p>\n<p><cite>Podle na\u0161eho n\u00e1zoru m\u016f\u017ee b\u00fdt c\u00edlen\u00ed protil\u00e1tek a jejich dod\u00e1v\u00e1n\u00ed do t\u011bla ve vysok\u00fdch koncentrac\u00edch \u00fa\u010dinnou n\u00e1hradou opakovan\u00fdch p\u0159eo\u010dkov\u00e1n\u00ed, zejm\u00e9na u rizikov\u00fdch skupin a lid\u00ed s oslaben\u00fdm imunitn\u00edm syst\u00e9mem.<\/cite><em>Infekce COVID-19 m\u016f\u017ee v\u00e9st k v\u00e1\u017en\u00e9mu onemocn\u011bn\u00ed a v\u00edme, \u017ee poskytnut\u00ed protil\u00e1tek v prvn\u00edch dnech po infekci m\u016f\u017ee zastavit \u0161\u00ed\u0159en\u00ed viru.\u00a0Je tedy mo\u017en\u00e9, \u017ee poskytnut\u00edm \u00fa\u010dinn\u00e9 protil\u00e1tkov\u00e9 l\u00e9\u010dby nebudeme muset poskytovat posilovac\u00ed d\u00e1vky cel\u00e9 populaci poka\u017ed\u00e9, kdy\u017e se objev\u00ed nov\u00e1 varianta.<\/em><\/p>\n<p class=\"has-medium-font-size\"><span style=\"font-size: 18pt;\"><strong>P\u0159irozen\u00e1 imunita je kl\u00ed\u010dov\u00e1<\/strong><\/span><\/p>\n<p>To je povzbudiv\u00e9, ne-li p\u0159\u00edmo p\u0159ekvapiv\u00e9, vzhledem k tomu, co je zn\u00e1mo o p\u0159irozen\u00e9 imunit\u011b \u2013 typu imunity z\u00edskan\u00e9 zotaven\u00edm z infekce.\u00a0Studie z roku 2022 publikovan\u00e1 v New England Journal of Medicine (NEJM) je jen jedn\u00edm z p\u0159\u00edklad\u016f v\u00fdzkumu,
