Dnes je nejv\u011bt\u0161\u00ed na sv\u011bt\u011b.<\/em><\/div>\n<\/div>\n<h2 id=\"Abs1\" class=\"c-article-section__title js-section-title js-c-reading-companion-sections-item\">Abstraktn\u00ed<\/h2>\n<div id=\"Abs1-content\" class=\"c-article-section__content\">\n<p>Cetylpyridiniumchlorid (CPC), kvart\u00e9rn\u00ed amoniov\u00e1 slou\u010denina, kter\u00e1 je p\u0159\u00edtomna v \u00fastn\u00ed vod\u011b, je \u00fa\u010dinn\u00e1 proti bakteri\u00edm, pl\u00edsn\u00edm a obalen\u00fdm vir\u016fm. Tato studie byla provedena s c\u00edlem prozkoumat antivirov\u00fd \u00fa\u010dinek CPC na SARS-CoV-2. Existuje n\u011bkolik zpr\u00e1v o \u00fa\u010dinku CPC proti SARS-CoV-2 divok\u00e9ho typu p\u0159i n\u00edzk\u00fdch koncentrac\u00edch, jako je 0,001 %\u20130,005 % (10\u201350 \u00b5g\/ml). Je zaj\u00edmav\u00e9, \u017ee jsme zjistili, \u017ee n\u00edzk\u00e9 koncentrace CPC potla\u010dily infek\u010dnost lidsk\u00fdch izolovan\u00fdch kmen\u016f SARS-CoV-2 (Wuhan, Alpha, Beta a Gamma) dokonce i ve slin\u00e1ch. D\u00e1le jsme pomoc\u00ed anal\u00fdzy hustoty sachar\u00f3zy a elektronov\u00e9ho mikroskopu prok\u00e1zali, \u017ee CPC vykazuje anti-SARS-CoV-2 \u00fa\u010dinky bez naru\u0161en\u00ed virov\u00e9ho obalu. SARS-CoV-2 S a N protein vykazoval specifickou distribuci v cel\u00e9m gradientu. Posun p\u00e1su byl pozorov\u00e1n u virion\u016f o\u0161et\u0159en\u00fdch Tritonem X-100; nicm\u00e9n\u011b frakce byly jin\u00e9 ne\u017e frakce o\u0161et\u0159en\u00e9 PBS nebo CPC. Jin\u00fdmi slovy, PBS a CPC nemus\u00ed m\u00edt \u017e\u00e1dn\u00fd vliv na strukturu virion\u016f SARS-CoV-2, zat\u00edmco Triton X-100 strukturu zm\u011bnil. D\u00e1le byla morfologie virion\u016f analyzov\u00e1na transmisn\u00ed elektronovou mikroskopi\u00ed (TEM). Elektronov\u00e1 mikroskopick\u00e1 anal\u00fdza odhalila, \u017ee struktura sf\u00e9rick\u00fdch \u010d\u00e1stic SARS-CoV-2 o\u0161et\u0159en\u00e9ho PBS z\u016fstala nezm\u011bn\u011bna. Tak\u00e9 jsme zjistili, \u017ee v\u011bt\u0161ina virov\u00fdch \u010d\u00e1stic o\u0161et\u0159en\u00fdch 10 ug\/ml CPC z\u016fstala nezm\u011bn\u011bna, zat\u00edmco n\u011bkter\u00e9 se rozpadly s 50 ug\/ml CPC. Naproti tomu t\u00e9m\u011b\u0159 v\u0161echny virov\u00e9 \u010d\u00e1stice o\u0161et\u0159en\u00e9 250 ug\/ml CPC byly jasn\u011b rozru\u0161eny (jako 1% Triton X-100). Mno\u017estv\u00ed 50 ug\/ml bylo pova\u017eov\u00e1no za koncentraci, p\u0159i kter\u00e9 se v\u0161echny virov\u00e9 \u010d\u00e1stice nerozbily. CPC prok\u00e1zal virucidn\u00ed \u00fa\u010dinky proti SARS-CoV-2 i ve slin\u00e1ch. Zjistili jsme tedy, \u017ee n\u00edzk\u00e9 koncentrace CPC vykazovaly dostate\u010dnou antivirovou aktivitu proti SARS-CoV-2. Proto\u017ee CPC naru\u0161uje lipidov\u00e9 dvojvrstvy, vysok\u00e9 koncentrace CPC mohou vykazovat cytotoxick\u00e9 \u00fa\u010dinky. Proto lze CPC aplikovat ve formulaci, kter\u00e1 m\u016f\u017ee uplat\u0148ovat sv\u016fj \u00fa\u010dinek po dlouhou dobu p\u0159i n\u00edzk\u00fdch koncentrac\u00edch. Proto\u017ee prototypov\u00e1 formulace \u00fastn\u00ed vody m\u016f\u017ee obsahovat dal\u0161\u00ed interferuj\u00edc\u00ed slo\u017eky, kter\u00e9 potla\u010duj\u00ed n\u00edzk\u00e9 hladiny CPC pou\u017eit\u00e9 v experimentech, testovali jsme komer\u010dn\u00ed \u00fastn\u00ed vodu se stejnou koncentrac\u00ed CPC,<a href=\"https:\/\/www-nature-com.translate.goog\/articles\/s41598-022-18367-6?_x_tr_sl=auto&amp;_x_tr_tl=cs&amp;_x_tr_hl=cs#MOESM1\" data-track=\"click\" data-track-label=\"link\" data-track-action=\"supplementary material anchor\">S2<\/a>\u00a0). T\u0159i nez\u00e1visl\u00e9 experimenty byly provedeny trojmo.<\/p>\n<h3 id=\"Sec13\" class=\"c-article__sub-heading\">Plakov\u00fd test<\/h3>\n<p>Kmeny SARS-CoV-2 byly sm\u00edch\u00e1ny se stejn\u00fdm mno\u017estv\u00edm roztoku CPC (kone\u010dn\u00e1 koncentrace: 0\u201350 \u00b5g\/ml s DMEM obsahuj\u00edc\u00edm 2 % FBS) nebo l\u00e9ka\u0159sk\u00e9ho kloktadla SP-T (Lion Corporation, Tokio, Japonsko), kter\u00e9 bylo na\u0159ed\u011bno. v PBS na koncentraci 50 ug\/ml, podobnou CPC. Sm\u011bs byla inkubov\u00e1na po dobu 30 minut p\u0159i teplot\u011b m\u00edstnosti a z\u0159ed\u011bna na 1\/10 2% FBS DMEM, aby se sn\u00ed\u017eil CPC ve sm\u011bsi. Z\u0159ed\u011bn\u00e1 sm\u011bs byla nao\u010dkov\u00e1na do bun\u011bk VeroE6\/TMPRSS2 a inkubov\u00e1na p\u0159i 37 \u00b0C po dobu 1 hodiny s rotac\u00ed. Po inkubaci byly bu\u0148ky dvakr\u00e1t promyty 1 x PBS, aby se odstranil CPC, a pot\u00e9 byly p\u0159ekryty 2% FBS DMEM obsahuj\u00edc\u00edm 1,2% Bacto Agar (Becton Dickinson, Franklin Lakes, NJ, USA). Po 48 hodin\u00e1ch inkubace p\u0159i 37 \u00b0C byly bu\u0148ky p\u0159es noc fixov\u00e1ny 3,7% pufrovan\u00fdm formaldehydem. Fixovan\u00e9 bu\u0148ky byly obarveny 1% krystalovou violet\u00ed.<\/p>\n<h3 id=\"Sec14\" class=\"c-article__sub-heading\">Test vstupu viru<\/h3>\n<p>Bu\u0148ky VeroE6\/TMPRSS2 byly nasazeny na 24jamkov\u00e9 desti\u010dky v hustot\u011b 1,0 x 105 bun\u011bk\/jamku\u00a0<sup>.\u00a0<\/sup>Kmen SARS-CoV-2 Wuhan byl sm\u00edch\u00e1n se stejn\u00fdm mno\u017estv\u00edm CPC (kone\u010dn\u00e1 koncentrace: 0\u201325 \u03bcg\/ml). P\u0159i ka\u017ed\u00e9 koncentraci l\u00e9\u010diva byly jamky infikov\u00e1ny 1,0 x\u00a0<sup>103<\/sup>PFU (MOI = 0,01) viru. Sm\u011bsi byly inkubov\u00e1ny po dobu 30 minut p\u0159i teplot\u011b m\u00edstnosti. Po inkubaci byly sm\u011bsi nao\u010dkov\u00e1ny do bun\u011bk VeroE6\/TMPRSS2 a inkubov\u00e1ny p\u0159i 37 \u00b0C po dobu 1 hodiny s rotac\u00ed. Po 1 hodin\u011b absorpce byly bu\u0148ky dvakr\u00e1t promyty 1 x PBS k odstran\u011bn\u00ed CPC a kultivov\u00e1ny v udr\u017eovac\u00edm m\u00e9diu. 24 hodin po infekci (hpi) byly celkov\u00e9 RNA extrahov\u00e1ny z nao\u010dkovan\u00fdch bun\u011bk pomoc\u00ed TRIzol\u2122 Reagent (Thermo Fisher Scientific, Waltham, MA, USA) a celkov\u00e1 RNA byla extrahov\u00e1na pomoc\u00ed RNeasy Mini Kit (QIAGEN, Hilden, N\u011bmecko). Extrahovan\u00e9 RNA byly podrobeny anal\u00fdze qRT-PCR pomoc\u00ed soupravy THUNDERBIRD Probe One-step qRT-PCR Kit (TOYOBO, Osaka, Japonsko). Genom SARS-CoV-2 byl kvantifikov\u00e1n pomoc\u00ed sad primerov\u00fdch sond pro N2 (Takara, Shiga, Japonsko). Jako endogenn\u00ed kontrola byl pou\u017eit \u03b2-aktin prim\u00e1t\u016f jin\u00e9ho ne\u017e lidsk\u00e9ho p\u016fvodu.<sup><a id=\"ref-link-section-d49693163e998\" title=\"Overbergh, L. a kol. TMPRSS11D a TMPRSS13 aktivuj\u00ed spike protein SARS-CoV-2.  Viry 13, 384 (2021).\" href=\"https:\/\/www-nature-com.translate.goog\/articles\/s41598-022-18367-6?_x_tr_sl=auto&amp;_x_tr_tl=cs&amp;_x_tr_hl=cs#ref-CR34\" data-track=\"click\" data-track-action=\"reference anchor\" data-track-label=\"link\" data-test=\"citation-ref\" aria-label=\"Odkaz 34\">\u00a0aktinu34<\/a><\/sup>\u00a0. Krom\u011b toho byly hladiny virov\u00e9 RNA p\u0159i 24 hpi normalizov\u00e1ny s hladinami virov\u00e9 RNA p\u0159i 0 hpi. V\u0161echny testy RT-PCR byly provedeny pomoc\u00ed syst\u00e9mu CFX96 Real-Time PCR System (BioRad, Hercules, CA, USA). T\u0159i nez\u00e1visl\u00e9 experimenty byly provedeny trojmo.<\/p>\n<h3 id=\"Sec15\" class=\"c-article__sub-heading\">Anal\u00fdza virucidn\u00ed aktivity CPC proti SARS-CoV-2 ve slin\u00e1ch<\/h3>\n<p>Kmen SARS-CoV-2 Wuhan byl p\u0159id\u00e1n do slin odebran\u00fdch od zdrav\u00e9ho dobrovoln\u00edka a sm\u00edch\u00e1n se stejn\u00fdm mno\u017estv\u00edm CPC (kone\u010dn\u00e1 koncentrace: 0\u201340 \u03bcg\/ml). Sm\u011bsi slin byly z\u0159ed\u011bny na 1\/100 pro sn\u00ed\u017een\u00ed viskozity a p\u0159efiltrov\u00e1ny p\u0159es 0,45 \u03bcm filtry (Sartorius) k odstran\u011bn\u00ed bakteri\u00ed a pl\u00edsn\u00ed. Plakov\u00fd test byl proveden tak, jak bylo pops\u00e1no d\u0159\u00edve (\u010d\u00e1st \u201e\u00a0<a href=\"https:\/\/www-nature-com.translate.goog\/articles\/s41598-022-18367-6?_x_tr_sl=auto&amp;_x_tr_tl=cs&amp;_x_tr_hl=cs#Sec13\" data-track=\"click\" data-track-label=\"link\" data-track-action=\"section anchor\">Plakov\u00fd test<\/a>\u00a0\u201c). T\u0159i nez\u00e1visl\u00e9 experimenty byly provedeny trojmo.<\/p>\n<h3 id=\"Sec16\" class=\"c-article__sub-heading\">Anal\u00fdza gradientu hustoty sachar\u00f3zy<\/h3>\n<p>Kmen SARS-CoV-2 Wuhan byl o\u0161et\u0159en CPC (50 \u03bcg\/ml) nebo Triton X-100 (1 %) p\u0159i pokojov\u00e9 teplot\u011b po dobu 10 minut. Po ultracentrifugaci s Optima XE-90 (Beckman Coulter, Brea, CA, USA) po dobu 6 hodin p\u0159i 250 000 \u00d7\u00a0<i>g<\/i>, ka\u017ed\u00fdch 100 \u03bcl gradient\u016f bylo frakcionov\u00e1no do 22 frakc\u00ed a sm\u00edch\u00e1no se 100 \u03bcl vzorkov\u00e9ho pufru SDS-PAGE a va\u0159eno p\u0159i 95 \u00b0C po dobu 5 minut. Po uva\u0159en\u00ed byly analyzov\u00e1ny pomoc\u00ed 10% SDS-PAGE s n\u00e1slednou imunoblotovou anal\u00fdzou s pou\u017eit\u00edm my\u0161\u00ed monoklon\u00e1ln\u00ed protil\u00e1tky proti SARS-CoV-2 S proteinu (GTX632604) nebo kr\u00e1li\u010d\u00edho polyklon\u00e1ln\u00edho N proteinu (GTX135357) (GeneTex, Irvine, CA, USA). Membr\u00e1ny byly po p\u0159enosu na\u0159\u00edznuty p\u0159i 100 kDa a hybridizov\u00e1ny s protil\u00e1tkou proti S proteinu, respektive s protil\u00e1tkou proti N proteinu a podrobeny vizualizaci. Pro zobrazov\u00e1n\u00ed byl pou\u017eit ImageQuant LAS 4000 mini (FUJIFILM Corporation, Tokio, Japonsko) (obr.\u00a0<a href=\"https:\/\/www-nature-com.translate.goog\/articles\/s41598-022-18367-6?_x_tr_sl=auto&amp;_x_tr_tl=cs&amp;_x_tr_hl=cs#MOESM1\" data-track=\"click\" data-track-label=\"link\" data-track-action=\"supplementary material anchor\">S4<\/a>\u00a0).<\/p>\n<h3 id=\"Sec17\" class=\"c-article__sub-heading\">Transmisn\u00ed elektronov\u00e1 mikroskopie<\/h3>\n<p>Kmen SARS-CoV-2 Wuhan byl o\u0161et\u0159en 1 x PBS, CPC (10, 50 a 250 ug\/ml) a Triton X-100 (1 %) po dobu 10 minut p\u0159i teplot\u011b m\u00edstnosti. Sm\u011bsi byly fixov\u00e1ny 2,5% glutaraldehydem p\u0159i 4 \u00b0C po dobu 24 hodin. Fixovan\u00fd vzorek (5 \u03bcl) byl um\u00edst\u011bn na list Parafilmu. Formvar Film (#10-1009, Okenshoji, Tokio, Japonsko) byl um\u00edst\u011bn na ka\u017edou kapku, aby adsorboval virus po dobu 5 minut. M\u0159\u00ed\u017eky byly promyty DDW a um\u00edst\u011bny na kapku filtrovan\u00e9ho 2,0% roztoku octanu uranylu na dal\u0161\u00ed 1 minutu, vysu\u0161eny na vzduchu a vy\u0161et\u0159eny pomoc\u00ed JEM-1400 TEM (JEOL, Tokyo, Japonsko) p\u0159i 80 kV.<\/p>\n<h3 id=\"Sec18\" class=\"c-article__sub-heading\">Statistika<\/h3>\n<p>Statistick\u00e9 anal\u00fdzy byly provedeny pomoc\u00ed Graphpad Prism v9 (GraphPad Software Inc., San Diego, CA, USA). Data jsou prezentov\u00e1na jako pr\u016fm\u011brn\u00e9 hodnoty \u00b1 SD biologick\u00fdch triplik\u00e1t\u016f. Statistick\u00e1 anal\u00fdza byla provedena pomoc\u00ed jednosm\u011brn\u00e9 anal\u00fdzy rozptylu.
